Supramolecular synthons in hydrates and solvates of lamotrigine: a tool for cocrystal design. [PDF]

open access: yesActa Crystallogr B Struct Sci Cryst Eng Mater
The molecule of anti-epileptic drug lamotrigine [LAM; 3,5-diamino-6-(2,3-dichlorophenyl)-1,2,4-triazine] is capable of the formation of multicomponent solids. Such an enhanced tendency is related to the diverse functionalities of the LAM chemical groups able to form hydrogen bonds.

1,1,2,2-Tetracyanocyclopropane (TCCP) as supramolecular synthon

open access: yesPhysical Chemistry Chemical Physics, 2016
The 1,1,2,2-tetracyanocyclopropane (TCCP) unit presents a synthetically accessible and versatile synthon that can interact with lone-pair or π-electrons by ‘non-covalent Carbon bonding’.

A Supramolecular Approach to Structure-Based Design with A Focus on Synthons Hierarchy in Ornithine-Derived Ligands: Review, Synthesis, Experimental and in Silico Studies

open access: yesMolecules, 2020
The success of innovative drugs depends on an interdisciplinary and holistic approach to their design and development. The supramolecular architecture of living systems is controlled by non-covalent interactions to a very large extent.

Modularity and three-dimensional isostructurality of novel synthons in sulfonamide–lactam cocrystals

open access: yesIUCrJ, 2015
The design of novel supramolecular synthons for functional groups relevant to drugs is an essential prerequisite for applying crystal engineering in the development of novel pharmaceutical cocrystals.

From a Binary to a Quaternary Cocrystal: An Unusual Supramolecular Synthon [PDF]

open access: yesAngewandte Chemie, 2019
AbstractFormation of a stoichiometric quaternary cocrystal consisting of resorcinol (RES), tetramethylpyrazine (TMP), phenazine (PHE) and pyrene (PYR) is described. A closed tetrameric resorcinol‐heterocycle synthon, unusual in that it has two different linker bases rather than just one, is observed in this four‐component solid.

Rapidly Diversifying Hydrogen‐Bonded Organic Frameworks With Permanent Porosity

open access: yesAngewandte Chemie, EarlyView.
This article reviews diversifying hydrogen‐bonded organic frameworks (HOFs) with permanent porosity reported over the past 15 years, based on the molecular structures of their constituent tectons. Furthermore, several distinctive aspects and concepts are described, such as permanent porosity, isostructural (isoreticular) HOFs, and multicomponent mixed ...

2-Pyridone: monoclinic polymorph

open access: yesActa Crystallographica Section E, 2009
The asymmetric unit in the title compound, C5H5NO, comprises two independent but virtually identical molecules of 2-pyridone, and represents a monoclinic polymorph of the previously reported orthorhombic (P212121) form [Penfold (1953). Acta Cryst. 6, 591&

Crystal Engineering of Ionic Cocrystals Sustained by Azolium···Azole Heterosynthons

open access: yesPharmaceutics, 2022
Crystal engineering of multi-component molecular crystals, cocrystals, is a subject of growing interest, thanks in part to the potential utility of pharmaceutical cocrystals as drug substances with improved properties. Whereas molecular cocrystals (MCCs)

Synthesis and Reactivity of New 1-Pentafluorophenyl-1Hpyrrole Derivatives

open access: yesMolecules, 2003
We present 1-pentafluorophenyl-1H-pyrrole and some of its electrophilic substitution reactions: formylation and acetylations. The 2-substituted products formed in these reactions are selectively and in high yield converted into 3-substituted products by ...

Supramolecular Synthon Identification in Azelaic Acid – Isonicotinamide

open access: yesJournal of Magnetic Resonance Open, 2022
Supramolecular synthons are the smallest structural units that contain the information inherent in building large molecular assemblies. Here, we apply solid-state NMR methods for the identification of the synthon in a cocrystal of Azelaic acid [(CH2)7(COOH)2]– Isonicotinamide [C6H6N2O]. It is observed that the spin-lattice relaxation time of protons in
