In vitro models of cancer‐associated fibroblast heterogeneity uncover subtype‐specific effects of CRISPR perturbations

open access: yesMolecular Oncology, EarlyView.
Development of therapies targeting cancer‐associated fibroblasts (CAFs) necessitates preclinical model systems that faithfully represent CAF–tumor biology. We established an in vitro coculture system of patient‐derived pancreatic CAFs and tumor cell lines and demonstrated its recapitulation of primary CAF–tumor biology with single‐cell transcriptomics ...

Detection of reflector surface error from near-field data: Effect of edge diffracted field [PDF]

open access: yes
The surface accuracy of large reflector antennas must be maintained within certain tolerances if high gain/low sidelobe performance is to be achieved. Thus the measurement of the surface profile is an important part of the quality control procedure when ...

Exploiting metabolic adaptations to overcome dabrafenib treatment resistance in melanoma cells

open access: yesMolecular Oncology, EarlyView.
We show that dabrafenib‐resistant melanoma cells undergo mitochondrial remodeling, leading to elevated respiration and ROS production balanced by stronger antioxidant defenses. This altered redox state promotes survival despite mitochondrial damage but renders resistant cells highly vulnerable to ROS‐inducing compounds such as PEITC, highlighting redox

Cell surface interactome analysis identifies TSPAN4 as a negative regulator of PD‐L1 in melanoma

open access: yesMolecular Oncology, EarlyView.
Using cell surface proximity biotinylation, we identified tetraspanin TSPAN4 within the PD‐L1 interactome of melanoma cells. TSPAN4 negatively regulates PD‐L1 expression and lateral mobility by limiting its interaction with CMTM6 and promoting PD‐L1 degradation.

PARP inhibition and pharmacological ascorbate demonstrate synergy in castration‐resistant prostate cancer

open access: yesMolecular Oncology, EarlyView.
Pharmacologic ascorbate (vitamin C) increases ROS, disrupts cellular metabolism, and induces DNA damage in CRPC cells. These effects sensitize tumors to PARP inhibition, producing synergistic growth suppression with olaparib in vitro and significantly delayed tumor progression in vivo. Pyruvate rescue confirms ROS‐dependent activity.

Relational Structure of Measurement with Application on Specification of Freeform Surface [PDF]

open access: yes, 2013
A contradiction is shown in this paper that, for contact surface measurement, if a measured surface profile is exactly coincident with the USL (upper specification limit), the measured result may still be out of specification.

Plecstatin inhibits hepatocellular carcinoma tumorigenesis and invasion through cytolinker plectin

open access: yesMolecular Oncology, EarlyView.
The ruthenium‐based metallodrug plecstatin exerts its anticancer effect in hepatocellular carcinoma (HCC) primarily through selective targeting of plectin. By disrupting plectin‐mediated cytoskeletal organization, plecstatin inhibits anchorage‐dependent growth, cell polarization, and tumor cell dissemination.

A Concept of the Effective Surface Profile to Predict the Roughness Parameters of Worn Surface

open access: yesFrontiers in Mechanical Engineering, 2019
A new concept of the effective surface profile is proposed to facilitate the prediction of the worn surface texture at sliding friction. The effective surface profile is a 2D height characteristic that consists of the asperities of surface superimposed ...

Polar Field Puzzle: Solutions from Flux-Transport Dynamo and Surface Transport Models

open access: yes, 2011
Polar fields in solar cycle 23 were about 50% weaker than those in cycle 22. The only theoretical models which have addressed this puzzle are surface transport models and flux-transport dynamo models.
