Targeting the HSPA8‐CMA‐ATP6V1A Axis Triggers Lysosomal Hyperacidification and Catastrophic Vacuolation in Prostate Cancer

Our experimental evidence supports a model in which ALO targets the HSPA8‐CMA‐ATP6V1A axis to induce lysosomal hyperacidification and initiate osmotic and lipidomic stress. These changes are associated with LMP and loss of lysosomal integrity in prostate cancer cells.

TMEM59L promotes neuroendocrine differentiation and enzalutamide resistance in prostate cancer by regulating MUC1/β‐catenin/MYC axis

Enzalutamide‐mediated AR suppression induces TMEM59L during prostate cancer progression. TMEM59L interacts with MUC1 and promotes its nuclear enrichment. Nuclear MUC1 activates β‐catenin/TCF4‐dependent MYC signalling, driving neuroendocrine differentiation, stemness and EMT.
