Evaluating Machine Learning Affinity Prediction Methods on Novel Lead Optimization Data: A 15‐PGDH Case Study

open access: yesChemMedChem, Volume 21, Issue 20, 28 October 2026.
Machine‐learning‐based protein–ligand affinity prediction has the potential to meaningfully impact drug discovery given their speed relative to traditional physics‐based methods. This case study considers recently disclosed inhibitors of 15‐hydroxyprostaglandin dehydrogenase and highlights the fragility of the domain of applicability of two methods ...

How Can the Latent Heat Entrainment Flux in the Convective Boundary Layer be Characterized?

open access: yesJournal of Geophysical Research: Atmospheres, Volume 131, Issue 20, 28 October 2026.
Abstract A proposed theoretical description of the latent heat entrainment flux at the top of a convective boundary layer (CBL) was tested. It is related to the vertical velocity scale times the ratio of the water vapor mixing ratio (WVMR) gradient to the Brunt‐Väisälä frequency. This relation was tested with different LiDAR and radiosonde measurements.

Synthesis, Moleculer Modeling, and Carbonic Anhydrase Inhibitor Effects of Novel 1‐(1‐benzyl‐2‐oxoindolin‐3‐ylidene)‐4‐(3‐sulfamoylphenyl)thiosemicarbazides

open access: yesChemMedChem, Volume 21, Issue 19, 14 October 2026.
Novel 1‐(1‐benzyl‐2‐oxoindolin‐3‐ylidene)thiosemicarbazides (6a–l) bearing a 3‐sulfamoylphenyl moiety selectively inhibited the tumor‐associated hCA IX and XII isoforms over hCA I and II isoforms at the nanomolar levels. Compounds 6k and 6f emerged as the most potent inhibitors of hCA IX and XII, respectively.
