Determination of thebaine in crude thebaine samples by infrared and ultraviolet spectrophotometric methods

The determination of the thebaine content of crude thebaine concentrates by infrared and ultraviolet spectrophotometric methods is described. The results obtained with the two methods were found to compare favourably. The accuracy of these procedures was found to be superior to that of the K–34 (U.N.) method.

Structural insights into thebaine synthase 2 catalysis

Thebaine synthase 2 (THS2) that can transform (7S)-salutaridinol 7-O-acetate to thebaine catalyzes the final step of thebaine biosynthesis in Papaver somniferum. Here, the crystal structures of THS2 and its complex with thebaine are reported, revealing the interaction network in the substrate-binding pocket.

Die Verbindung ist stereoisomer mit dem schon bekannten „Thebainon‐enol‐methyläther”︁ (V) und unterscheidet sich von diesem durch Konfigurationsänderung am C‐Atom 14.

Cyclo-adducts of thebaine with nitrosoarenes

Thebaine (1) reacts reversibly with various nitroso-arenes to form cyclo-adducts by 1,4-addition of the nitrosogroup to the conjugated diene system of the alkaloid. Adducts (2) of thebaine with nitrosobenzene and with 4-chloro-, 4-methyl-, 3-methoxy-, and 4-nitro-nitrosobenzene have been prepared in good yield.
