Dearomative Access to (−)-Thebaine and Derivatives

Organic Letters, 2023
A synthesis of the natural product thebaine is reported in eight steps from commercially available starting materials, hinging on the dearomatization and coupling of simple aromatic starting materials. This provides divergent access to two unnatural opioid derivatives and is aimed at the long-term development of synthetic opioid analogs of the ...

Structural insights into thebaine synthase 2 catalysis

Biochemical and Biophysical Research Communications, 2020
Thebaine synthase 2 (THS2) that can transform (7S)-salutaridinol 7-O-acetate to thebaine catalyzes the final step of thebaine biosynthesis in Papaver somniferum. Here, the crystal structures of THS2 and its complex with thebaine are reported, revealing the interaction network in the substrate-binding pocket.

A cluster of acute thebaine poisonings from non-food grade poppy seeds in the Australian food supply

Clinical toxicology, 2023
Introduction Poppy seed tea is used for its opioid effects and contains multiple opium alkaloids, including morphine, codeine, papaverine, and thebaine.

Toxicokinetics of thebaine in those consuming non-food grade poppy seeds as a tea

Clinical toxicology, 2023
Introduction Thebaine is an alkaloid in poppy seeds that is neurotoxic to animals. Data on its clinical effects and toxicokinetics in people are minimal.

An industrially applicable Escherichia coli platform for bioconversion of thebaine to oripavine and codeine to morphine.

Chemical Communications, 2023
A whole cell Escherichia coli biotransformation platform converting thebaine to oripavine and codeine to morphine was demonstrated with industrially applicable yields (∼1.2 × 10-2 g L-1 h-1 or ∼1.2 × 10-1 g L-1 h-1), improving >13 400-fold upon morphine ...

A Seven-Step Total Synthesis of (–)-Thebaine

Synthesis, 2022
The morphinan alkaloid (–)-thebaine (1) is an industrially important chemical intermediate deployed in the semi-synthesis of various opioid medicines.

Urine Thebaine Determination by Liquid Chromatography Tandem Mass Spectrometry After Poppy Seed Consumption.

Drug Testing and Analysis, 2022
Laboratories are challenged to distinguish whether a positive urine morphine result is due to heroin use or possible poppy seed consumption. Thebaine is an opium alkaloid that has been shown to be present in the urine of individuals who have consumed ...
