The development and applications of polyclonal and monoclonal antibodies for the detection of illicit drugs in saliva samples [PDF]

open access: yes, 2002
Anti-tetrahydrocannabinol (THC), anti-cocaine and anti-morphine polyclonal antibodies were produced. These antibodies were successfully applied to an ELISA format for the detection of THC, cocaine, and morphine in saliva samples. Monoclonal antibodies

High-Affinity Nanobody Against the LEDGF PWWP Domain Inhibits Chromatin Binding In Vitro

open access: yesBiomolecules
Background and objectives: The PWWP domain of lens epithelium-derived growth factor p75 (LEDGF/p75) mediates chromatin engagement through recognition of histone H3 lysine 36 di- and trimethylation (H3K36me2/3) and nucleosomal DNA.

Tau acetylation at K331 has limited impact on tau pathology in vivo

open access: yesFEBS Letters, EarlyView.
We mapped tau post‐translational modifications in humanized MAPT knock‐in mice and in amyloid‐bearing double knock‐in mice. Acetylation within the repeat domain, particularly around K331, showed modest increases under amyloid pathology. To test functional relevance, we generated MAPTK331Q knock‐in mice.

Advancements in the application of reporter gene cell lines in bioactivity evaluation of biological products

open access: yesBioresources and Bioprocessing
The assessment of biological product activity is a key aspect of quality control. Currently, in vitro assays serve as the primarily method employed by both companies and regulatory agencies to evaluate biological activity.

Calpain small subunit homodimerization is robust and calcium‐independent

open access: yesFEBS Letters, EarlyView.
Calpains dimerize via penta‐EF‐hand (PEF) domains. Using single‐molecule force spectroscopy, we measured the strength and kinetics of PEF–PEF homodimer binding. The interaction is robust, shows a transient conformational step before dissociation, and remains largely insensitive to Ca2+.

Antibody modulation: Limiting the efficacy of therapeutic antibodies [PDF]

open access: yesPharmacological Research, 2015
Monoclonal antibodies (mAb) have revolutionised the way in which we treat disease. From cancer to autoimmunity, antibody therapy has been responsible for some of the most impressive clinical responses observed in the last 2 decades. A key component of this success has been their generally low levels of toxicity, and unique mechanisms of action.

Gut microbiome and aging—A dynamic interplay of microbes, metabolites, and the immune system

open access: yesFEBS Letters, EarlyView.
Age‐dependent shifts in microbial communities engender shifts in microbial metabolite profiles. These in turn drive shifts in barrier surface permeability of the gut and brain and induce immune activation. When paired with preexisting age‐related chronic inflammation this increases the risk of neuroinflammation and neurodegenerative diseases.

High-concentration L-methionine as a potent antioxidant for oxidation resistance and stability enhancement in high-concentration antibody therapeutics

open access: yesInternational Journal of Pharmaceutics: X
The development of high-concentration monoclonal antibody (mAb) formulations for subcutaneous administration is faces critical stability challenges, particularly oxidation and aggregation, which compromise efficacy and safety.
