Assaying activity and assessing thermostability of hyperthermophilic enzymes [PDF]
There is now a wide variety of intra- and extra-cellular enzymes available from organisms growing above 75°C, and having sufficient stability to allow assay well above this temperature.

Moorella thermoacetica is the model organism for elucidation of acetogenesis. The reduction of CO2 to acetate is well studied, but how redox carrier balancing and energy conservation are maintained during autotrophic growth and how H2 is oxidised remains enigmatic.

The functional potential of microbial communities in hydraulic fracturing source water and produced water from natural gas extraction characterized by metagenomic sequencing [PDF]
Microbial activity in produced water from hydraulic fracturing operations can lead to undesired environmental impacts and increase gas production costs. However, the metabolic profile of these microbial communities is not well understood.

Characterization of ferredoxins from the thermophilic, acetogenic bacterium Thermoanaerobacter kivui
A major electron carrier involved in energy and carbon metabolism in the acetogenic model organism Thermoanaerobacter kivui is ferredoxin, an iron–sulfur‐containing, electron‐transferring protein. Here, we show that the genome of T. kivui encodes four putative ferredoxin‐like proteins (TKV_c09620, TKV_c16450, TKV_c10420 and TKV_c19530).

A (2R,3R)‐butanediol dehydrogenase from Bacillus subtilis (BsBDH) is engineered for the enantioselective synthesis of 2‐hydroxycyclohexanone. A PASS computational design strategy is proposed to enhance the thermostability of BsBDH. Moreover, ESM‐1v combined with ISM is utilized for enhancing and inverting its stereoselectivity.

Two novel thermophilic bacteria, Thermincola strain AZ34E and Carboxydocella strain AZ29I, were isolated from hydrothermal environments and shown to oxidise carbon monoxide via the water‐gas shift reaction. Comparative genomic and phylogenetic analyses revealed that all currently described isolates within Thermincola and Carboxydocella each represent a

Enrichment and microbial characterization of syngas converting anaerobic cultures [PDF]
Bioconversion of recalcitrant biomass/waste into bulk chemicals or biofuels is often not feasible. By gasification of these materials, syngas (mainly composed of CO2, CO and H2) is generated and can be used for the production of high value compounds by ...

Biomass‐Derived Diformylxylose as a Renewable Solvent for Biocatalysis Applications
It feels greener in diformylxylose (DFX)! The renewable solvent dissolves aromatic ketones at remarkable concentrations and sustains the performance of robust enzymes such as ketoreductases (KREDs) and lipases. By combining superior substrate dissolution with improved enzyme performance, DFX offers a green medium for selective, high‐load ...

Gene Encoding a Trehalose Phosphorylase fromThermoanaerobacter brockiiATCC 35047 [PDF]
A gene encoding a trehalose phosphorylase was cloned from Thermoanaerobacter brockii ATCC 35047. The gene encodes a polypeptide of 774 amino acid residues. The deduced amino acid sequence was homologous to bacterial maltose phosphorylases and a trehalose 6-phosphate phosphorylase catalyzing anomer-inverting reactions. On the other hand, no homology was

Careful CO Addition Enhances Autotrophic d‐Lactate Formation With Engineered Acetobacterium woodii
ABSTRACT The acetogen A. woodii efficiently converts CO2 and H2 to acetate. Metabolic engineering enabled the autotrophic production of non‐native products, for example, d‐lactate from CO2 by overexpression of d‐lactate dehydrogenase from Leuconostoc mesenteroides and knockout of the native lactate dehydrogenase. During gas fermentation with acetogens,
