Expression of thiopurine methyltransferase in South Asians [PDF]
Azathioprine and its metabolite 6-mercaptopurine (6-MP) are widely used in the treatment of Crohn’s disease, ulcerative colitis, autoimmune hepatitis, and a range of other inflammatory and autoimmune diseases, as well as for prevention of transplant rejection.

BACKGROUND: Azathioprine (AZA), used to treat inflammatory bowel disease (IBD), is metabolized by thiopurine methyltransferase (TPMT). The accumulation of individual metabolites varies because humans display genetic polymorphism for TPMT expression ...

Clinical Pharmacogenetics Implementation Consortium Guidelines for Thiopurine Methyltransferase Genotype and Thiopurine Dosing [PDF]
Thiopurine methyltransferase (TPMT) activity exhibits monogenic co-dominant inheritance, with ethnic differences in the frequency of occurrence of variant alleles. With conventional thiopurine doses, homozygous TPMT-deficient patients (~1 in 178 to 1 in 3,736 individuals with two nonfunctional TPMT alleles) experience severe myelosuppression, 30-60% of

Personalization of thiopurine therapy: Current recommendations and future perspectives
Despite great therapeutic advances in the field of biologics, small synthetic molecules such as thiopurines, including azathioprine, mercaptopurine, and thioguanine, remain an important therapeutic pillar in the treatment of inflammatory bowel disease ...

Direct evidence of singlet molecular oxygen [O2 (1Δg)] production from UVA excited 6‐thioguanine
6‐Thioguanine (6‐TGua) is incorporated into DNA as a purine analogue, inhibiting cell replication. Patients treated with 6‐TGua are more prone to developing skin cancer due to the photoexcitation of 6‐TGua by UVA radiation (as illustrated in the Jablonski diagram). Upon exposure to UVA, the excited 6‐TGua generates 1O2.

6-methylmercaptopurine-induced leukocytopenia during thiopurine therapy in inflammatory bowel disease patients [PDF]
Background and Aim: Thiopurines have a favorable benefit–risk ratio in the treatment of inflammatory bowel disease. A feared adverse event of thiopurine therapy is myelotoxicity, mostly occurring due to toxic concentrations of the pharmacologically ...

Genotyping of Thiopurine Methyltransferase Using Pyrosequencing
Thiopurine methyltransferase (TPMT) metabolizes thiopurine drugs which are used in the treatment of leukemia and some autoimmune diseases. Previously, 11 mutant alleles of TPMT gene (TPMT*1S, *2, *3A, *3B, *3C, *3D, *4, *5, *6, *7, and *8) have been reported.

Case Report: Azathioprine: An Old and Wronged Immunosuppressant
Mycophenolate rapidly substituted azathioprine (AZA) in transplant immunosuppression regimens since the 1990s, when early clinical trials indicated better outcomes, although opposite results were also observed. However, none of these trials used the well-
