The hypothermic effect of neurotensin (0.1 microgram) injected i.c.v. in mice is potentiated by the enkephalinase inhibitor thiorphan (10 micrograms, i.c.v.). This potentiation is not reversed by systemic naloxone. The hypothermic effect of neurotensin is not modified by the amino-peptidase inhibitor bestatin (50 micrograms, i.c.v.) nor by the ...

The effect of inhaled thiorphan on allergen-induced airway responses in asthmatic subjects
Clinical <html_ent glyph="@amp;" ascii="&"/> Experimental Allergy, 1996SummaryBackground Neuropeptides are likely to be implicated in the pathophysiology of allergen‐induced airway responses. However, upon release in the airways, neuropeptides are potentially inactivated by neutral endopeptidase (NEP). Objective We hypothesized that NEP‐inhibition by inhaled thiorphan (TH) would increase allergen‐induced early (EAR)

Effect of bestatin and thiorphan on [Met5]enkephalin-Arg6-Phe7-induced analgesia
European Journal of Pharmacology, 1984We investigated the effect of bestatin, a specific inhibitor of aminopeptidase and thiorphan, a specific inhibitor of enkephalinase A, on the analgesic effect induced by the intracerebral injection of heptapeptide [Met5]enkephalin-Arg6-Phe7 (MEAP) in cannulated rats.

Potentiation of the hypoglycemic effect of insulin by thiorphan, an enkephalinase inhibitor
European Journal of Pharmacology, 1984Insulin administration to rats produced a dose-related hypoglycemia. When insulin and the enkephalinase (Enk'ase) inhibitor thiorphan (30 or 100 mg/kg s.c.) were co-administered, there was a potentiation of the hypoglycemic response to insulin; these doses of thiorphan alone had no significant effect on plasma glucose.

Substance P-induced effects in guinea pig lungs: effects of thiorphan and captopril
Journal of Applied Physiology, 1989The effects of the neutral metalloendopeptidase inhibitor, thiorphan, and the angiotensin-converting enzyme inhibitor, captopril, on the changes in airway opening pressure (PaO), pulmonary arterial pressure (Ppa), and weight induced by intravascular administration of substance P were examined in isolated perfused and ventilated guinea pig lungs ...

Antitumor activity of bestatin and thiorphan in mice.
Archivum immunologiae et therapiae experimentalis, 1996We have investigated the effect of bestatin and thiorphan on growth of murine transplantable tumors, survival time, activity of natural killer (NK) cells and macrophages in mice. The injections of thiorphan at the does of 0.5 or 5 micrograms per mouse retarded tumor growth and prolonged survival period in B16 melanoma bearing animals. Pretreatment with

The peptidase inhibitors thiorphan and bestatin were tested for their ability to inhibit the actions of the oligopeptidases contained in the venom of the Taiwan cobra (Naja naja atra). With methionine enkephalin (TyrGlyGlyPheMet) as substrate, thiorphan was an effective inhibitor of cleavage of the GlyPhe peptide bond while bestatin inhibited cleavage ...

Disposition of thiorphan in DOCA-salt and spontaneously hypertensive rats.
Research communications in chemical pathology and pharmacology, 1993Thiorphan was administered intravenously (i.v.) at 10 mg/kg to conscious rats in two different models of hypertension to allow a comparison of pharmacokinetics. The two models were: 1) Deoxycorticosterone acetate (DOCA)-salt uninephrectomized rats; 2) Spontaneously hypertensive rats (SHR), and their respective normotensive controls; 3) Sprague-Dawley ...
