IMPDH inhibition enhances cytarabine efficacy in SAMHD1‐expressing leukaemia cells via guanine nucleotide depletion

open access: yesMolecular Oncology, EarlyView.
Cytarabine is a key therapy for acute myeloid leukaemia (AML), but its efficacy is limited by the dNTPase SAMHD1, which hydrolyses its active metabolite. Screening nucleotide biosynthesis inhibitors revealed that IMPDH inhibitors selectively sensitise SAMHD1‐proficient AML cells to cytarabine.

Time Series Representation for Visualization in Apache IoTDB

open access: yesProceedings of the ACM on Management of Data
When analyzing time series, often interactively, the analysts frequently demand to visualize instantly large-scale data stored in databases. M4 visualization selects the first, last, bottom and top data points in each pixel column to ensure pixel-perfectness of the two-color line chart visualization.

Keratin 19 as a prognostic marker and contributing factor of metastasis and chemoresistance in high‐grade serous ovarian cancer

open access: yesMolecular Oncology, EarlyView.
Keratin 19 (KRT19) is overexpressed in high‐grade serous ovarian cancer with high levels of Kallikrein‐related peptidases (KLK) 4–7 and is associated with poor survival. In vivo analyses demonstrate that elevated KRT19 increases peritoneal tumour burden.

Surrogate-Assisted Symbolic Time-Series Discretization Using Multi-Breakpoints and a Multi-Objective Evolutionary Algorithm

open access: yesMathematical and Computational Applications
The enhanced multi-objective symbolic discretization for time series (eMODiTS) method employs a flexible discretization scheme using different value cuts for each non-equal time interval, which incurs a high computational cost for evaluating each ...

Dual PI3K/AKT and CDK4/6 inhibition reveals selective sensitivity in an SHH medulloblastoma stem cell model

open access: yesMolecular Oncology, EarlyView.
Targeted therapy was evaluated in SHH medulloblastoma using neuroepithelial stem cell (NES) and tumor‐derived NES‐like (tNES) models in 2D monolayers and 3D spheroids. PI3K, AKT, and CDK4/6 inhibitors had minimal effects in NES but markedly reduced viability and growth and induced apoptosis in tNES cells, revealing distinct therapeutic vulnerabilities.

Time Series Anomaly Detection Leveraging MSE Feedback with AutoEncoder and RNN [PDF]

open access: yes
Anomaly detection in time series data is a critical task in various domains, including finance, healthcare, cybersecurity and industry. Traditional methods, such as time series decomposition, clustering, and density estimation, have provided robust ...

Scalable Univariate and Multivariate Time-Series Classifiers with Deep Learning Methods Exploiting Symbolic Representations

open access: yesComputers
Time-series classification (TSC) is an important task across sciences. Symbolic representations (especially SFA) are very effective at combating noise. In this paper, we employ symbolic representations to create state-of-the-art time-series classifiers ...

Establishment of a humanized patient‐derived xenograft mouse model of high‐grade serous ovarian cancer for preclinical evaluation of combination immunotherapy

open access: yesMolecular Oncology, EarlyView.
We have established a humanized orthotopic patient‐derived xenograft (Hu‐oPDX) mouse model of high‐grade serous ovarian cancer (HGSOC) that recapitulates human tumor–immune interactions. Using combined anti‐PD‐L1/anti‐CD73 immunotherapy, we demonstrate the model's improved biological relevance and enhanced translational value for preclinical ...

Deciphering transcriptional plasticity in pancreatic ductal adenocarcinoma reveals alterations in sensory neuron innervation

open access: yesMolecular Oncology, EarlyView.
Pancreatic sensory neurons innervating healthy and PDAC tissue were retrogradely labeled and profiled by single‐cell RNA sequencing. Tumor‐associated innervation showed a dominant neurofilament‐positive subtype, altered mitochondrial gene signatures, and reduced non‐peptidergic neurons.

MCPL-Based FT-LSTM: Medical Representation Learning-Based Clinical Prediction Model for Time Series Events

open access: yesIEEE Access, 2019
Large collections of electronic medical records (EMRs) provide us with a vast source of information on medical practice. However, the utilization of these data to support clinical decisions is still limited.
