Immediate Growth Control in Response to Boiling Histotripsy is Prognostic for Intratumoral Immune Activation

open access: yesAdvanced Science, EarlyView.
Boiling Histrotripsy (BH), a mechanical form of focused ultrasound (FUS), allows for non‐invasive and non‐ionizing tissue destruction. Utilizing a BH treatment protocol that yielded bifurcating primary tumor growth responses, we found that immediate BH‐induced tumor growth control identifies tumors with acute reductions in antigen‐presenting cells (APC)

Mannosylated T/Tn with Freund’s adjuvant induces cellular immunity

open access: yesInternational Journal of Immunopathology and Pharmacology, 2018
Inducing cancer-specific cellular immune responses has become an attractive strategy in cancer treatment. In this study, we investigated the role of several adjuvants in eliciting T/Tn-specific cellular immunity and protection against T/Tn expressing ...

Repurposing a Small Molecule Plant Hormone as a Tunable ON‐Switch for CAR‐T Cell Immunotherapy

open access: yesAdvanced Science, EarlyView.
By engineering a receptor system integrating the plant auxin receptor AFB1 with its co‐receptor IAA7, we enable ligand‐dependent interactions triggered by the plant hormone auxins. This design allows rapid, reversible, and dose‐dependent T cell activation, resulting in potent cytotoxicity against B‐cell lymphoma in vitro and in vivo.

Tn-Core: A Toolbox for Integrating Tn-seq Gene Essentiality Data and Constraint-Based Metabolic Modeling

open access: yes, 2018
The design of synthetic cells requires a detailed understanding of the relevance of genes and gene networks underlying complex cellular phenotypes. Transposon-sequencing (Tn-seq) and constraint-based metabolic modeling can be used to probe the core ...

An Innervated Human Skin Equivalent that Electrically Encodes Mechanical and Thermal Stimuli

open access: yesAdvanced Science, EarlyView.
An innervated human skin equivalent is integrated with high‐density microelectrode arrays to reveal how engineered free nerve ending–like structures translate touch and temperature into electrical codes. Mechanical indentation and thermal stimulation engage the engineered somatosensory circuitry and evoke distinct firing and waveform signatures ...

Truncated O-glycosylation in metastatic triple-negative breast cancer reveals a gene expression signature associated with extracellular matrix and proteolysis

open access: yesScientific Reports
Breast cancer (BC) is the leading cause of death by cancer in women worldwide. Triple-negative (TN) BC constitutes aggressive and highly metastatic tumors associated with shorter overall survival of patients compared to other BC subtypes. The Tn antigen,

Natural anti-Tn-reactive IgM (cross-reactive anti-A isoagglutinin) is neutralized by blood group A phenotype- determining glycosylation, as evident in SARS-CoV-2 (COVID-19) infection

open access: yes, 2023
Exposure of a human host to SARS-CoV-2 results in activation of the host trans-membrane protease serine subtype 2 (TMPRSS2) and the ontogenetic polypeptide GalNAc transferase, resulting in a cross-species, mucin-type O-glycan bridge, the classical Tn ...

A Drug‐Gated, Modular STAb‐T Immunotherapy With External Control

open access: yesAdvanced Science, EarlyView.
Engineered STAb‐TON cells act as programmable living factories that continuously secrete inactive antibody modules, which assemble extracellularly into a functional T cell engager only upon small‐molecule administration. This externally controlled platform enables reversible, on‐demand regulation of antitumor immunity, establishing a new paradigm for ...
