A fully synthetic four‐component vaccine has been developed combining Tn antigen, vizantin (Mincle agonist), rhamnose (antibody recruitment), and mannose (CD206 targeting). By synergistically activating Mincle, FcγR, and CD206 pathways, it elicits strong IgG switching and cytotoxic T‐cells, significantly inhibiting tumors in mice.

Glucose deprivation in the primary CNS lymphoma (PCNSL) tumor microenvironment drives SLC2A5 (encoding GLUT5)‐dependent fructose metabolism in tumor cells, while hypoxia induces HIF‐mediated SLC2A5 expression in tumor‐supportive macrophages, revealing SLC2A5‐driven fructose utilization as a shared and targetable metabolic vulnerability across malignant

The engineered DTSe‐based photosensitizer, DZTSe, emerges as an avant‐garde molecular entity, which can distribute to both the endoplasmic reticulum and mitochondria to concurrently trigger pyroptosis and activate the cGAS‐STING pathway. This dual‐action strategy effectively reprograms the tumor microenvironment, enabling potent eradication of primary ...

Are pancarcinoma T and Tn differentiation antigens? [PDF]
Human fetal tissues were studied for presence of immunoreactive Thomsen-Friedenreich (T) and Tn epitopes (EPs) using well-defined anti-T and anti-Tn rodent monoclonal antibodies. T and Tn are universal (pan) carcinoma (CA) markers that are occluded in normal postfetal tissues except in some immunoprivileged enclaves.

Biodegradable Acoustic Targeting for Ultrasound‐Supported Gene Therapy (BATUS) in Glioblastoma
ABSTRACT Glioblastoma (GBM) remains one of the most challenging brain malignancies due to the restrictive nature of the blood–brain barrier (BBB), which severely limits effective drug and gene delivery. To overcome this, we introduce Biodegradable Acoustic Targeting for Ultrasound‐Supported Gene Therapy (BATUS), a modular platform that enables safe ...

Summary: The Tn antigen is a well-known tumor-associated carbohydrate determinant, often incorporated in glycopeptides to develop cancer vaccines. Herein, four copies of a conformationally constrained mimetic of the antigen TnThr (GalNAc-Thr) were ...

Glycan elongation beyond the mucin associated Tn antigen protects tumor cells from immune-mediated killing. [PDF]
Membrane bound mucins are up-regulated and aberrantly glycosylated during malignant transformation in many cancer cells. This results in a negatively charged glycoprotein coat which may protect cancer cells from immune surveillance. However, only limited
