Epigenetic heterogeneity and plasticity in therapy‐induced tumor states through single‐cell multi‐omics

open access: yesMolecular Oncology, EarlyView.
Single‐cell multi‐omics reveals epigenetic heterogeneity across therapy‐adaptive tumor states, including quiescent/dormant, drug‐tolerant persister, and EMT‐like phenotypes. By linking regulatory features with state‐associated biomarkers, these approaches inform biomarker‐guided therapeutic strategies for evolving tumors.

Cutting tool condition monitoring of the turning process using artificial intelligence

open access: yes, 2012
This thesis relates to the application of Artificial Intelligence to tool wear monitoring. The main objective is to develop an intelligent condition monitoring system able to detect when a cutting tool is worn out.

In vitro and in silico modelling of ROS1‐positive non‐small cell lung cancer reveals fusion‐dependent tyrosine kinase inhibitor responses

open access: yesMolecular Oncology, EarlyView.
Drug resistance limits treatment success in a subset of lung cancers driven by ROS1 gene alterations. Using patient‐derived cells and computer simulations, we studied three key mutations and how they affect five targeted drugs. The mutations reduced drug effectiveness in different ways by altering protein structure and behavior.

NT-proBNP as a risk stratification tool for the management of acute decompensated heart failure [PDF]

open access: yes, 2018
The burden of heart failure (HF) remains high, with worrisome morbidity and mortality rates for patients hospitalized for acute decompensated heart failure (ADHF). The natriuretic peptides, BNP and the inactive N-terminal fragment of its precursor proBNP

ZW4864‐mediated inhibition of the β‐catenin/BCL9/BCL9L complex reveals therapeutic potential in bladder cancer

open access: yesMolecular Oncology, EarlyView.
BCL9 and BCL9L drive bladder cancer progression by enhancing β‐catenin signaling, promoting proliferation, migration, invasion, and organoid growth. Genetic depletion of BCL9(L) suppresses malignant phenotypes, while pharmacological disruption of the β‐catenin/BCL9(L) complex with ZW4864 inhibits canonical Wnt signaling and tumor‐associated cellular ...

Analysing the significance of small conformational changes and low occupancy states in serial crystallographic data

open access: yesFEBS Open Bio, EarlyView.
This protocol paper outlines methods to establish the success of a time‐resolved serial crystallographic experiment, by means of statistical analysis of timepoint data in reciprocal space and models in real space. We show how to amplify the signal from excited states to visualise structural changes in successful experiments.

Proteasomal degradation of intracellularly expressed Amblyomin‐X limits suicide gene therapy potential in melanoma cells

open access: yesFEBS Open Bio, EarlyView.
This study explores the feasibility of expressing the antitumoral protein Amblyomin‐X through a suicide gene therapy approach and investigates its intracellular fate after gene delivery. Although the gene is efficiently expressed, melanoma cells rapidly degrade the Amblyomin‐X protein via proteasome activity.

CETGI : an assessment tool for global concurrent engineering : executive summary [PDF]

open access: yes
Industrial globalisation and the increased complexity of products promote new product development (NPD) by distributed teams. These teams facilitate the transfer of designs to manufacturing facilities, the adaptation of products to local markets and ...

Delayed failure of software components using stochastic testing [PDF]

open access: yes, 2012
The present research investigates the delayed failure of software components and addresses the problem that the conventional approach to software testing is unlikely to reveal this type of failure. Delayed failure is defined as a failure that occurs some

COFUN: A new tool for evaluating containment failure frequency and uncertainty

open access: yesNuclear Engineering and Technology
The feature of the COntainment Failure probability and Uncertainty aNalysis program (COFUN) is the quantification of the containment state. The probability factors used in probabilistic safety assessment (PSA) and the resulting outputs contain ...
