Induction of diplochromosomes in mammalian cells by inhibitors of topoisomerase II

Chromosoma, 1998
Diplochromosomes, consisting of four chromatids lying side-by-side, instead of the normal two, are produced when cells go through two rounds of DNA replication without separation of chromatids. They are thus an indication of the failure of the normal chromosome separation mechanism.

Genotoxicity of topoisomerase II inhibitors: An anti-infective perspective

Toxicology, 2008
At present, an inevitable consequence of a chemical's inhibitory activity on key regulators of DNA topology in bacteria, the type II topoisomerases, is a less pronounced effect on their eukaryotic counterparts. In the context of anti-infectives drug development, this may pose a risk to patient safety as inhibition of eukaryotic type II topoisomerases ...

[Molecular determinants of response to topoisomerase I inhibitors].

Bulletin du cancer, 2011
Nuclear topoisomerase I (Top1) is involved in the relaxation of DNA supercoiling and plays a pivotal role in the coordination of essential DNA processes such as transcription, replication, DNA recombination and DNA damage signalling. For all these reasons, Top1 has been an attractive target for the development of anticancer drugs, which poison Top1 by ...

DNA topoisomerase: the mechanism of resistance to DNA topoisomerase II inhibitor VP-16. [PDF]

open access: possibleHiroshima journal of medical sciences, 1990
K6-1 and 50B-3 cell lines, resistant to VP-16, a DNA topoisomerase II inhibitor, were established from two different types of cells respectively: human T-cell derived acute lymphoblastic leukemia cell line RPMI8402 and mouse mammary tumor cell line FM3A.
