TR146 cells grown on filters as a model of human buccal epithelium: V. Enzyme activity of the TR146 cell culture model, human buccal epithelium and porcine buccal epithelium, and permeability of leu-enkephalin

open access: yesInternational Journal of Pharmaceutics, 2000
The objective of the present study was to characterise the TR146 cell culture model as an in vitro model of human buccal mucosa with respect to the enzyme activity in the tissues. For this purpose, the contents of aminopeptidase, carboxypeptidase and esterase in homogenate supernatants of the TR146 cell culture model, and human and porcine buccal ...

TR146 cells grown on filters as a model of human buccal epithelium: permeability of fluorescein isothiocyanate-labelled dextrans in the presence of sodium glycocholate

open access: yesJournal of Controlled Release, 1999
The aim of the present study was to characterize the TR146 cell culture model as an in vitro model of human buccal epithelium with respect to the permeability of test substances with different molecular weights (M(w)). For this purpose, the apparent permeability (P(app)) values for mannitol and for fluorescein isothiocyanate (FITC)-labelled dextrans ...

TR146 cells grown on filters as a model of human buccal epithelium: III. Permeability enhancement by different pH values, different osmolality values, and bile salts

open access: yesInternational Journal of Pharmaceutics, 1999
The aim of the present study was to evaluate the TR146 cell culture model as an in vitro model of human buccal epithelium with respect to the permeability enhancement by different pH values, different osmolality values or bile salts. For this purpose, the increase in the apparent permeability (P(app)) of the hydrophilic marker mannitol due to exposure ...

The Potential of Chitosan in Enhancing Peptide and Protein Absorption Across the TR146 Cell Culture Model—An in Vitro Model of the Buccal Epithelium

open access: yesPharmaceutical Research, 2002
To investigate the potential of chitosan (CS) to enhance buccal peptide and protein absorption, the TR146 cell culture model, a model of the buccal epithelium, was used. The sensitivity of TR146 cells to several CS solutions (different salts with different MW) was investigated by using the MTS/ PMS assay. Permeability studies were performed to determine

Filter-grown TR146 cells as an in vitro model of human buccal epithelial permeability

European Journal of Oral Sciences, 1999
Use of a cell culture model of a specific epithelium requires documentation of its differentiation. This study reports permeability of mannitol concurrent with a profile of differentiation markers of filter-grown TR146 cells, a cell line originating from a human buccal carcinoma, cultivated submerged or at the air-liquid interface for 23 to 31 d.

TR146 cells grown on filters as a model for human buccal epithelium: I. Morphology, growth, barrier properties, and permeability

International Journal of Pharmaceutics, 1995
Abstract The characterization and functional application of filter-grown TR146 cells are presented. TR146 is a continued cell line of human buccal epithelial origin. Growth parameters indicated a saturation density in the order of 2 × 105 cells/cm2 and a doubling time ranging from 33 to 49 h dependent on type of support.

TR146 cells as a model for human buccal epithelium: II. Optimisation and use of a cellular sensitivity MTS/PMS assay

International Journal of Pharmaceutics, 1996
Abstract The optimisation and use of the cellular sensitivity MTS/PMS assay for TR146 cells are presented. The MTS/PMS assay is a colorimetric method based on reduction of a tetrazolium salt to the corresponding formazan including an electron acceptor as accelerator.

Cyclodextrin inclusion complexes of antimycotics intended to act in the oral cavity – drug supersaturation, toxicity on TR146 cells and release from a delivery system

European Journal of Pharmaceutics and Biopharmaceutics, 1999
The dissolution rate, the toxicity and the release from chewing gum of miconazole and econazole cyclodextrin products and complexes were investigated. The dissolution rate studies showed that an amorphous miconazole hydroxypropyl-beta-cyclodextrin product gave drug supersaturation, whereas drug supersaturation was not present during dissolution rate ...

The membrane-associated MUC1 improves adhesion of salivary MUC5B on buccal cells. Application to development of an in vitro cellular model of oral epithelium

open access: yesArchives of Oral Biology, 2016
Objectives: The mucosal pellicle is a thin layer of salivary proteins, mostly MUC5B mucins, anchored to epithelial oral cells. This pellicle is involved in protection of oral mucosae against abrasion, pathogenic microorganisms or chemical xenobiotics ...
