[13C]cholesterol as a tracer for studies of cholesterol metabolism in humans

open access: yesJournal of Lipid Research, 1993
The investigation of cholesterol metabolism in humans would be facilitated by the availability of a tracer that is not radioactive. However, to be useful such a tracer must be detectable in the large pool of body cholesterol over an extended time.

A light‐triggered Time‐Resolved X‐ray Solution Scattering (TR‐XSS) workflow with application to protein conformational dynamics

open access: yesFEBS Open Bio, EarlyView.
Time‐resolved X‐ray solution scattering captures how proteins change shape in real time under near‐native conditions. This article presents a practical workflow for light‐triggered TR‐XSS experiments, from data collection to structural refinement. Using a calcium‐transporting membrane protein as an example, the approach can be broadly applied to study ...

Promiscuous stimulation of HSP70 ATPase activity by parasite‐derived J‐domains

open access: yesFEBS Open Bio, EarlyView.
The malaria parasite Plasmodium falciparum exports three highly homologous yet functionally divergent J‐domain proteins into human erythrocytes. Here, we show that J‐domains isolated from all three proteins effectively stimulate the ATPase activity of both endogenous host and exported parasite HSP70 chaperones.

Samples Preparation of Breath Test 14C Tracer for Accelerator Mass Spectrometry

open access: yesJournal of Isotopes, 2017
Carbon plays an important role in the metabolism of plants and human. Radiocarbon (14C) is an ideal tracer for vivo human metabolism, disease diagnosis and organ function assessment studies.

The Gravitational Potential Near the Sun From SEGUE K-dwarf Kinematics

open access: yes, 2013
To constrain the Galactic gravitational potential near the Sun ($\sim$1.5 kpc), we derive and model the spatial and velocity distribution for a sample of 9000 K-dwarfs that have spectra from SDSS/SEGUE, which yield radial velocities and abundances ([Fe/H]

Single‐molecule DNA flow‐stretch assays for high‐throughput DNA–protein interaction studies

open access: yesFEBS Open Bio, EarlyView.
We describe an optimised single‐molecule DNA flow‐stretch assay that visualises DNA–protein interactions in real time. Linear DNA fragments are tethered to a surface and stretched by buffer flow for fluorescence imaging. Using λ and φX174 DNA, this protocol enhances reproducibility and accessibility, providing a versatile approach for studying diverse ...

DSi as a Tracer for Submarine Groundwater Discharge

open access: yesFrontiers in Marine Science, 2019
Submarine groundwater discharge (SGD) is an important source of nutrients and metals to the coastal ocean, affects coastal ecosystems, and is gaining recognition as a relevant water resource.

Optimizing photoactivation of PA‐mCherry for optical pooled CRISPR screens

open access: yesFEBS Open Bio, EarlyView.
Photoactivatable PA‐mCherry finds widespread use to optically tag individual cells. However, confocal 405 nm UV laser‐scanning (normal scan) is much less efficient than widefield UV illumination, limiting the use of PA‐mCherry on confocal instruments. We remedy this limitation by reporting that rapid and repeated confocal scanning with a low‐intensity,

Sudden anaerobization in Amphibacillus xylanus increases intracellular labile ferrous iron and inhibits cell growth

open access: yesFEBS Open Bio, EarlyView.
Abruptly changing from aerobic to anaerobic conditions (sudden anaerobization) induced growth inhibition and a significant increase in intracellular labile ferrous iron in the aerotolerant anaerobe Amphibacillus xylanus. We found that free flavins mediate efficient electron transfer from NADH to ferric iron under anaerobic conditions, suggesting that ...

Aquaporin-4-dependent glymphatic solute transport in the rodent brain

open access: yeseLife, 2018
The glymphatic system is a brain-wide clearance pathway; its impairment contributes to the accumulation of amyloid-β. Influx of cerebrospinal fluid (CSF) depends upon the expression and perivascular localization of the astroglial water channel aquaporin ...
