Structural Studies on Transfer RNA: Crystallization of Formylmethionine and Leucine Transfer RNA's

Science, 1969
Improved solvent systems were used to crystallize two different transfer RNA species. These crystals show increased mechanical and thermal stability over crystals obtained previously from a similar system. They have sufficient stability and crystalline order to be used in x-ray crystallographic studies.

The role of cations (polyamines and Mg2+) in isoleucyl-tRNA formation catalyzed by purified isolecuyl-tRNA synthetase [EC 6.1.1.5] from Escherichia coli was studied. It was found that spermine, spermidine, and Mg2+ bind to tRNA and that when bound to these cations, tRNA acts as substrate of aminoacylation without requiring further cations.

On the origin of the transfer RNA molecule

Data and arguments are given in favour of the hypothesis that the primitive tRNA molecule may have originated from a direct duplication event involving one of the two halves of the tRNA molecule. It seems that a molecule capable of assuming a hairpin structure was involved as a precursor in this duplication.
