Icariin Enhances the Enzymatic Activity of N‐acetylgalactosaminidase to Augment Akkermansia Abundance in Gut Microbiota for Improved PD‐1 Blockade Efficacy in Tumor Suppression

open access: yesAdvanced Science, EarlyView.
Icariin promoted the growth of Akk by enhancing the activity of N‐acetylgalactosaminidase (Amuc_0920), which enhanced mucin utilization and provided a favorable nutrient environment for bacterial growth. This icariin‐mediated enrichment of Akk further reshaped the tumor microenvironment and promoted CD8+ T cell infiltration, ultimately synergizing with

Corticosteroids impair epithelial regeneration in immune-mediated intestinal damage

open access: yesThe Journal of Clinical Investigation
Corticosteroid treatment (CST) failure is associated with poor outcomes for patients with gastrointestinal (GI) graft-versus-host disease (GVHD). CST is intended to target the immune system, but the glucocorticoid receptor (GR) is widely expressed ...

A Microbial Lipid‐ATP Synthase Axis Fuels NK Cell Antitumor Activity

open access: yesAdvanced Science, EarlyView.
This study focuses on the mechanism by which gut microbiota‐derived outer membrane vesicles (OMVs) regulate NK cell antitumor activity. B. intestinalis is identified to decrease extra‐intestinal tumor growth via its OMVs enriched in sphingosine (SP).

B cells mediate lung ischemia/reperfusion injury by recruiting classical monocytes via synergistic B cell receptor/TLR4 signaling

open access: yesThe Journal of Clinical Investigation
Ischemia/reperfusion injury–mediated (IRI-mediated) primary graft dysfunction (PGD) adversely affects both short- and long-term outcomes after lung transplantation, a procedure that remains the only treatment option for patients suffering from end-stage ...

RENAL‐CHIP: Rejection Evaluation via Non‐Invasive Analysis of Circulating Podocytes With Herringbone‐Chip Isolation Platform

open access: yesAdvanced Science, EarlyView.
RENAL‐CHIP converts 1 mL of peripheral blood into a biopsy‐equivalent readout of renal‐allograft fate. By magnetic capture and release of donor‐derived circulating podocytes through a herringbone microfluidic chip, 84% capture, 96% release and single‐cell RNA evidence of rejection‐specific immunity are achieved.

Treg activation during allograft tolerance induction requires mitochondrion-induced TGF-β1 in type 1 conventional dendritic cells

open access: yesThe Journal of Clinical Investigation
The role of conventional type 1 DCs (cDC1s) in tolerance induction to solid organ allografts is unknown and important for strategies that seek to prolong allograft viability. Using a murine model deficient in cDC1s, we report cDC1s are required for donor

AML‐Targeted Metal‐Polyphenol Nanoplatform Induces Ferroptosis‐ICD Cascade for Antitumor Immunity Boosting

open access: yesAdvanced Science, EarlyView.
Schematic illustration of the synthesis protocol for Fe‐SH@Fn, an AML‐targeted nanoplatform engineered to co‐deliver shikonin (SH) and Fe3+, synergistically boosting antitumor immunity via ferroptosis induction and immunogenic cell death (ICD). ABSTRACT Acute myeloid leukemia (AML) remains a therapeutic challenge due to its low immunogenicity and ...

Calcineurin inhibition rescues alloantigen-specific central memory T cell subsets that promote chronic GVHD

open access: yesThe Journal of Clinical Investigation
Calcineurin inhibitors (CNIs) constitute the backbone of modern acute graft-versus-host disease (aGVHD) prophylaxis regimens but have limited efficacy in the prevention and treatment of chronic GVHD (cGVHD).

T Cell Exhaustion in Cancer Immunotherapy: Heterogeneity, Mechanisms, and Therapeutic Opportunities

open access: yesAdvanced Science, EarlyView.
T cell exhaustion limits immunotherapy efficacy. This article delineates its progression from stem‐like to terminally exhausted states, governed by persistent antigen, transcription factors, epigenetics, and metabolism. It maps the exhaustion landscape in the TME and proposes integrated reversal strategies, providing a translational roadmap to overcome
