Tropane alkaloid biosynthesis: a centennial review

Natural Product Reports, 2021
From the first ambitious imagination of tropinone biosynthesis mechanism in plants published in 1917 to the de novo production of scopolamine in yeast realized in 2020, what did we learn from this long story of more than 100 years old?

Fluorinated Tropanes

Current Topics in Medicinal Chemistry, 2013
The development of tropane-derivatives as cocaine-addiction therapeutics identified numerous compounds with affinities for the dopamine, serotonin, and norepinephrine transporters. Many of these compounds were then adapted for use as imaging radiotracers.

The Distorted Tropane of Scopoline

ChemPhysChem, 2013
AbstractThe structural isomerization of scopine into scopoline (oscine) has been observed in a supersonic jet expansion using microwave spectroscopy. The rotational spectrum evidences a single structure in the gas phase, providing a first description of the (three‐ring) structurally distorted tropane in scopoline.

The conformations of tropanes

Journal of the Chemical Society C: Organic, 1966
A combination of proton-resonance-spectroscopic and dipole-moment measurements confirms that tropanes exist with the piperidine ring in a chair conformation and with the N-methyl substituent predominantly equatorial. A 3α-substituent causes considerable ring deformation.
