Prospects for non-immunological molecular therapeutics in melanoma [PDF]

open access: yes, 2010
In 2006 there were 60,000 new cases of cutaneous melanoma in the European Union and 13,000 deaths (www.europeancancerleagues.org). Currently available systemic treatment options for metastatic melanoma, including both cytotoxic and immunologic therapies,

A novel quinazolinone insulin receptor inhibitor and its synergy with an EGFR inhibitor in glucose‐driven glioblastoma

open access: yesMolecular Oncology, EarlyView.
The novel styrylquinazolinone‐based molecule W1B effectively suppresses glioblastoma by inhibiting IGF1R and EGFR. In high‐glucose microenvironments driving tumor resistance, W1B acts synergistically with the EGFR inhibitor dacomitinib. This combination safely blocks compensatory survival signaling in zebrafish xenograft models. Showcasing promising in

Latest progress in tyrosine kinase inhibitors

open access: yesOncotarget, 2014
Here we discuss the latest progress in development of some kinase inhibitors such as inhibitors of c-MET, LIM and Bcr-Abl kinases. Importantly, many oncogenic kinases signal via the mTOR pathway, suggesting a common target for drug combinations.

Structural and functional characterization of the intracellular kinase domain of vascular endothelial growth factor receptor-2 [PDF]

open access: yes, 2013
Vascular Endothelial Growth Factors (VEGFs) are key players in blood and lymphatic vessel development and homeostasis. The family consists of five members, VEGF-A, -B, -C, -D and placenta growth factor (PLGF).

Epigenetic heterogeneity and plasticity in therapy‐induced tumor states through single‐cell multi‐omics

open access: yesMolecular Oncology, EarlyView.
Single‐cell multi‐omics reveals epigenetic heterogeneity across therapy‐adaptive tumor states, including quiescent/dormant, drug‐tolerant persister, and EMT‐like phenotypes. By linking regulatory features with state‐associated biomarkers, these approaches inform biomarker‐guided therapeutic strategies for evolving tumors.

Untersuchungen zur Regulierung und Funktion der Mitogen-aktivierten Proteinkinase ERK5 = Regulation and Function of the Mitogen-activated Protein Kinase ERK5

open access: yes, 2003
Mitogen activated protein kinases (MAPKs) are found in all eukaryotic cells and represent crucial elements in the signal transduction from the plasma membrane to the nucleus.

Combinatorial strategy using protein kinase inhibitors and a cytotoxic compound for highly resistant glioblastoma cells : "in vitro studies" [PDF]

open access: yes, 2007
Glioblastoma multiforme (GBM) is the most frequent and the most aggesssive malignant neoplasm of the human central nervous system (CNS), with a median survival of less than one year. These neoplasms are radio- and chemo-resistant, and are highly invasive,

PAK1 activation drives divergent resistance mechanisms to aromatase inhibition and tamoxifen in a luminal: A breast cancer model

open access: yesMolecular Oncology, EarlyView.
Breast cancer remains a major cause of cancer death in women, frequently developing endocrine therapy resistance. This study demonstrates that upregulated p21‐activated kinase 1 (PAK1) activity drives resistance to tamoxifen and long‐term estrogen deprivation in ER+ breast cancer models.

Mechanism of Na+/Ca2+ Exchanger Activation by Hydrogen Peroxide in Guinea-Pig Ventricular Myocytes

open access: yesJournal of Pharmacological Sciences, 2007
Using the whole-cell voltage clamp, we examined the mechanism of activation of the Na+/Ca2+ exchanger (NCX) by hydrogen peroxide (H2O2) in isolated guinea-pig cardiac ventricular myocytes. Exposure to H2O2 increased the NCX current.

Management of imatinib-resistant CML patients [PDF]

open access: yes, 2007
Imatinib has had marked impact on outcomes in chronic myelogenous leukemia (CML) patients for all stages of the disease and is endorsed by international treatment guidelines as the first line option.
