A Ubiquitin-Conjugating Enzyme Is Essential for Developmental Transitions inDictyostelium [PDF]
We have identified a developmentally essential gene,UbcB, by insertional mutagenesis. The encoded protein (UBC1) shows very high amino acid sequence identity to ubiquitin-conjugating enzymes from other organisms, suggesting that UBC1 is involved in protein ubiquitination and possibly degradation during Dictyostelium development.

Summary: Ubiquitin and ubiquitin-like chains are finely balanced by conjugating and de-conjugating enzymes. Alterations in this balance trigger the response to stress conditions and are often observed in pathologies.

Robust cullin-RING ligase function is established by a multiplicity of poly-ubiquitylation pathways
The cullin-RING ligases (CRLs) form the major family of E3 ubiquitin ligases. The prototypic CRLs in yeast, called SCF enzymes, employ a single E2 enzyme, Cdc34, to build poly-ubiquitin chains required for degradation. In contrast, six different human E2

A quantitative assay to monitor HSV-1 ICP0 ubiquitin ligase activity in vitro [PDF]
The ubiquitin–proteasome system is an essential cellular process that plays a fundamental role in the regulation of protein stability. This pathway is tightly controlled by a sequential cascade of enzymatic steps that culminates in the formation of a ...

Post‐Translational Modifications in Cilia and Ciliopathies
This review synthesizes current understanding of post‐translational modifications (PTMs) in ciliary proteins and emphasizes their roles in ciliary formation, homeostasis, and signaling. This review also discusses the implication of PTM dysregulation in ciliopathies and explores therapeutic strategies targeting PTM‐modifying enzymes.

Three-dimensional structure of a ubiquitin-conjugating enzyme (E2).
The x-ray crystal structure of a recombinant ubiquitin-conjugating enzyme (E2) encoded by the UBC1 gene of the plant Arabidopsis thaliana has been determined with the use of multiple isomorphous replacement techniques and refined at 2.4-A resolution by simulated annealing and restrained least-squares.

Discovery of cellular regulation by protein degradation [PDF]
What follows is a story of some of the lab’s adventures mentioned above, including the inventions of new biochemical and genetic methods. This account stems, in part, from previous descriptions of the early history of the Ub field (31,32).

This study elucidates the critical role of Rictor in macrophage activation in acute antibody‐mediated rejection (ABMR). Rictor increases K48‐linked ubiquitination of p65 by upregulating E3 ubiquitin ligase SOCS1, inhibiting transcriptional levels of NLRP3 and inflammasome activation.

Spatiotemporal Regulation of STING Activity by Linear Ubiquitination Governs Antiviral Immunity
This study uncovers a spatiotemporal regulation of STING activity by linear ubiquitination in antiviral immunity. At the early stage of the infection, LUBAC promotes STING linear ubiquitination to drive its trafficking from the ER to the Golgi apparatus for activation.
