Role of RING-Type E3 Ubiquitin Ligases in Inflammatory Signalling and Inflammatory Bowel Disease

open access: yesMediators of Inflammation, 2020
Ubiquitination is a three-step enzymatic cascade for posttranslational protein modification. It includes the ubiquitin-activating enzyme (E1), ubiquitin-conjugating enzyme (E2), and ubiquitin ligase (E3).

Anti‐Inflammatory Mechanisms of Selenium Nanosheets in Ulcerative Colitis: Protein Corona, GP130 Interaction, and Transcriptomic Profile

open access: yesAdvanced Science, EarlyView.
SeNSs provide a biocompatible, anti‐inflammatory UC therapy. SeNSs form protein coronas enriched with AKT/PI3K/NF‐κB pathway proteins, suppress GP130 via hydrophobic interactions, and inhibit pro‐inflammatory cytokines. In DSS‐induced UC mice, SeNSs reduce inflammation, tissue damage, and disease activity by modulating cytokine, chemokine, and ...

Optimized protocol to detect protein UFMylation in cells and in vitro via immunoblotting

open access: yesSTAR Protocols, 2022
Summary: Ubiquitin-fold modifier 1 (UFM1) system is a recently identified ubiquitin-like modification with essential biological functions. Similar to ubiquitination, the covalent conjugation of UFM1 (UFMylation) to target proteins involves a three-step ...

Nephronectin (NPNT) is a Crucial Determinant of Idiopathic Pulmonary Fibrosis: Modulating Cellular Senescence via the ITGA3/YAP1 Signaling Axis

open access: yesAdvanced Science, EarlyView.
Through a comprehensive multi‐omics analysis, this study identifies a marked reduction in Nephronectin (NPNT) expression within fibrotic lung tissue. This reduction impairs the binding capability to the ITGA3 receptor, consequently causing YAP1 to persist in the cytoplasm, where it undergoes degradation.

TRIM21‐Mediated K11‐Linked Ubiquitination of ID1 Suppresses Tumorigenesis and Promotes Cuproptosis in Esophageal Squamous Cell Carcinoma

open access: yesAdvanced Science, EarlyView.
ID1 is regulated by E3 ligase TRIM21, which adds non‐degradative K11‐linked ubiquitin at Lys91, disrupting ID1‐TCF12 binding and freeing TCF12 to upregulate copper transporter SLC31A1. Enhanced SLC31A1 elevates intracellular copper, triggers cuproptosis, and inhibits ESCC growth.

EBBP‐Mediated Integrated Stress Response Attenuates Anthracycline‐Induced Cardiotoxicity by Inhibiting the Ferroptosis of Cardiomyocytes

open access: yesAdvanced Science, EarlyView.
EBBP expression is significantly upregulated in DOX‐treated cardiomyocytes. Mechanistically, EBBP interacts with GRP78 to mediate K63‐linked ubiquitination, thereby attenuating the inhibitory GRP78‐PERK interaction and triggering activation of the PERK‐mediated integrated stress response (ISR).

Combined Genetic Association and Differed Expression Analysis of UBE2L3 Uncovers a Genetic Regulatory Role of (Immuno)proteasome in IgA Nephropathy

open access: yesKidney Diseases
Introduction: IgA nephropathy (IgAN) is a leading cause of end-stage renal disease. The exact pathogenesis of IgAN is not well defined, but some genetic studies have led to a novel discovery that the (immuno)proteasome probably plays an important role in

LncDARS‐AS1 Regulates ATP1A1 Stability and Enhances Na+/K+ ATPase Activity to Promote Osteosarcoma Metastasis

open access: yesAdvanced Science, EarlyView.
This study identifies LncDARS‐AS1 as a novel oncogenic regulator that stabilizes ATP1A1 and enhances Na⁺/K⁺ ATPase activity, promoting osteosarcoma metastasis. Targeting the LncDARS‐AS1/ATP1A1/NKA axis, including pharmacological inhibition with digoxin, offers promising opportunities for the development of new therapeutic strategies against ...
