UFL1, a UFMylation E3 ligase, plays a crucial role in multiple cellular stress responses

open access: yesFrontiers in Endocrinology, 2023
The UFM1 conjugation system(UFMylation)is a novel type of ubiquitin-like system that plays an indispensable role in maintaining cell homeostasis under various cellular stress.

Ubiquitination of transcription factors in cancer: unveiling therapeutic potential

open access: yesMolecular Oncology, EarlyView.
In cancer, dysregulated ubiquitination of transcription factors contributes to the uncontrolled growth and survival characteristics of tumors. Tumor suppressors are degraded by aberrant ubiquitination, or oncogenic transcription factors gain stability through ubiquitination, thereby promoting tumorigenesis.

The E2 Ubiquitin-conjugating Enzyme UBE2J1 Is Required for Spermiogenesis in Mice [PDF]

open access: yesJournal of Biological Chemistry, 2014
ER-resident proteins destined for degradation are dislocated into the cytosol by components of the ER quality control machinery for proteasomal degradation. Dislocation substrates are ubiquitylated in the cytosol by E2 ubiquitin-conjugating/E3 ligase complexes. UBE2J1 is one of the well-characterized E2 enzymes that participate in this process. However,

Targeted protein degradation in oncology: novel therapeutic opportunity for solid tumours?

open access: yesMolecular Oncology, EarlyView.
Current anticancer therapies are limited by the occurrence of resistance and undruggability of most proteins. Targeted protein degraders are novel, promising agents that trigger the selective degradation of previously undruggable proteins through the recruitment of the ubiquitin–proteasome machinery. Their mechanism of action raises exciting challenges,

Conformational Dynamics Modulate Activation of the Ubiquitin Conjugating Enzyme Ube2g2 [PDF]

open access: yesACS Omega, 2017
The ubiquitin conjugating enzyme Ube2g2 together with its cognate E3 ligase gp78 catalyzes the synthesis of lysine-48 polyubiquitin chains constituting signals for the proteasomal degradation of misfolded proteins in the endoplasmic reticulum. Here, we employ NMR spectroscopy in combination with single-turnover diubiquitin formation assays to examine ...

Ubiquitin-conjugating enzyme E2C (UBE2C) is a prognostic indicator for cholangiocarcinoma

open access: yesEuropean Journal of Medical Research, 2023
AbstractCholangiocarcinoma is the most common malignant bile duct tumor in Southeast Asia. The special location of cholangiocarcinoma leads to it being difficult to diagnose. Currently, the progress in clinical prognosis outcomes remains abysmal owing to the lack of definitive diagnostic criteria.

Small modifier, big decision: switching to SUMO mode adds weight to cancer stemness in mammary tumors

open access: yesMolecular Oncology, EarlyView.
Inhibition of protein SUMOylation has been shown to block tumorigenesis; however, the specific mechanisms by which SUMOylation controls the tumor‐initiating capacities remain elusive. Li et al. describe the role of Etv1 SUMOylation in cancer stem cells using mouse models of mammary gland tumorigenesis.

Regulation of lysosome integrity and lysophagy by the ubiquitin-conjugating enzyme UBE2QL1 [PDF]

open access: yesAutophagy, 2019
Lysosomal membrane permeabilization or full rupture of lysosomes is a common and severe stress condition that is relevant for degenerative disease, infection and cancer. Cells respond with extensive ubiquitination of damaged lysosomes, which triggers selective macroautophagy/autophagy of the whole organelle, termed lysophagy.

Liver‐specific lncRNAs associated with liver cancers

open access: yesFEBS Open Bio, EarlyView.
Long non‐coding RNAs (lncRNAs) are regulatory molecules with various functions. They are more tissue‐specific than proteins and can be used as potential biomarkers, particularly in cancer diagnostics and prognosis. In this review, we have systematically compiled all lncRNAs with exclusive expression in the human liver, verified their liver specificity ...
