Regulation of biological processes by ubiquitin ligases: a focus on the Pagano Lab’s contribution

open access: yesFrontiers in Cell and Developmental Biology
Protein homeostasis depends on many fundamental processes including mRNA synthesis, translation, post-translational modifications, and proteolysis.

Calhm6 Governs Macrophage Polarization Through Chp1‐Camk4‐Creb1 Axis and Ectosomal Delivery in Inflammatory Responses

open access: yesAdvanced Science, EarlyView.
Calhm6 drives M2 macrophage polarization via the Chp1‐Camk4‐Creb1 axis, suppressing inflammation through calcium‐dependent ectosomal delivery. Calhm6 deficiency enhances M1 responses, boosting bactericidal activity but exacerbating tissue damage. LPS/IFNγ upregulate Calhm6 via Irf1, while IL‐4/Stat6 inhibits it, balancing immune outcomes.

Adenovirus Protein Involved in Virus Internalization Recruits Ubiquitin−Protein Ligases

open access: yesBiochemistry, 2002
Adenovirus penton base protein is involved in virus internalization. Searching for the cellular partners of this protein, we used dodecahedra, adenovirus subviral particles composed of 12 bases, for screening a human lung expression library. This screen yielded three ubiquitin-protein ligases, WWP1, WWP2, and AIP4, all of which belong to the HECT ...

Dual‐Targeted Nanovesicles Induced Cancer Stem‐Like Cell Differentiation to Sensitize Hepatocellular Carcinoma Radiotherapy

open access: yesAdvanced Science, EarlyView.
The existence of Pin1 and Notch1 crosstalk is identified and verified in HCC, and the crucial function of this crosstalk in sustaining the aggressive phenotypes of CSCs and radioresistance. A biomimetic nanovesicle is now developed to block the crosstalk between Pin1 and Notch1 for inducing CSCs differentiation and sensitizing HCC radiotherapy ...

A Blast‐Resistant NLR Gene Confers Drought Resistance by Competitively Interacting with an E3 Ligase to Protect Phenylalanine Ammonia‐Lyase in Rice

open access: yesAdvanced Science, EarlyView.
The NLR protein PibH8 interacts with phenylalanine ammonia‐lyase OsPAL1 and E3 ubiquitin ligase OsFBK16. Through competitive binding, PibH8 weakens the interaction between OsFBK16 and OsPAL1, thereby reducing the degradation of OsPAL1 by OsFBK16. The Hap1 allele of PibH8 exhibits a higher expression level than Hap2, thus accumulating more PibH8 protein,

Proteasome Inhibitors: Harnessing Proteostasis to Combat Disease [PDF]

open access: yes, 2020
The proteasome is the central component of the main cellular protein degradation pathway. During the past four decades, the critical function of the proteasome in numerous physiological processes has been revealed, and proteasome activity has been linked

Mesencephalic Astrocyte‐Derived Neurotrophic Factor Binds BAX to Preserve Mitochondrial Homeostasis and Energy Metabolism for Relieving Myocardial Hypertrophy

open access: yesAdvanced Science, EarlyView.
MANF in cardiomyocytes interacts with BAX to impede BAX mitochondrial translocation and cytochrome c release, further stabilizing mitochondrial structure and energy supply to relieve myocardial apoptosis and hypertrophy, which indicates MANF as a novel cardioprotective factor to have a promising clinical application for myocardial hypertrophy ...
