Structure-Guided Design and Optimization of Small Molecules Targeting the Protein–Protein Interaction between the von Hippel–Lindau (VHL) E3 Ubiquitin Ligase and the Hypoxia Inducible Factor (HIF) Alpha Subunit with in Vitro Nanomolar Affinities

open access: yesJournal of Medicinal Chemistry, 2014
E3 ubiquitin ligases are attractive targets in the ubiquitin–proteasome system, however, the development of small-molecule ligands has been rewarded with limited success.

HSV-1 ICP0: An E3 Ubiquitin Ligase That Counteracts Host Intrinsic and Innate Immunity

open access: yesCells, 2014
The herpes simplex virus type 1 (HSV-1) encoded E3 ubiquitin ligase, infected cell protein 0 (ICP0), is required for efficient lytic viral replication and regulates the switch between the lytic and latent states of HSV-1.

METTL10‐Mediated PIAS3 Methylation Links Purine Metabolism to Gastric Cancer Progression

open access: yesAdvanced Science, EarlyView.
METTL10‐mediated methylation of PIAS3 at K442 inhibits its SUMO E3 ligase activity toward MITF. This reduces MITF SUMOylation and subsequent RNF4‐mediated ubiquitination, leading to MITF protein accumulation and gastric cancer progression. Abstract Metabolic dysregulation plays a significant role in the development of gastric cancer (GC).

YKT6 Promotes Bladder Cancer Progression by Stabilizing β‐catenin Through USP7‐Mediated Deubiquitination

open access: yesAdvanced Science, EarlyView.
The SNARE protein YKT6 is upregulated in bladder cancer (BLCA), correlating with poor prognosis. YKT6 promotes tumor proliferation and metastasis by activating Wnt/β‐catenin signaling. It recruits Ubiquitin‐Specific Peptidase 7 (USP7) to deubiquitinate and stabilize β‐catenin, enhancing its nuclear accumulation and driving oncogenic gene expression ...
