Characterization of Ubrogepant: A Potent and Selective Antagonist of the Human Calcitonin Gene‒Related Peptide Receptor [PDF]
A growing body of evidence has implicated the calcitonin gene-related peptide (CGRP) receptors in migraine pathophysiology. With the recent approval of monoclonal antibodies targeting CGRP or the CGRP receptor, the inhibition of CGRP-mediated signaling has emerged as a promising approach for preventive treatments of migraine in adults.

Migraine: Epidemiology, Risk Factors, Pathophysiology, and Treatment
Migraine is a highly prevalent, female‐predominant, and disabling brain disorder shaped by genetic susceptibility, hormonal influences, modifiable attack factors, and comorbidity‐related progression. This review integrates current evidence on migraine epidemiology, phase‐specific neurovascular and neuroglial mechanisms, including CGRP signaling and ...

Abstract A population pharmacokinetic (popPK) model was developed for ubrogepant using data from 10 Phase 1, 1 Phase 2, and 2 Phase 3 studies. The data were described by a 2‐compartment model with linear elimination and transit compartment absorption. Formulation, food intake, race, gender, and hepatic impairment had a statistically significant impact ...

Cerebrospinal Fluid and Plasma Concentrations of Atogepant in Humans
ABSTRACT The aim of this study was to determine the cerebrospinal fluid (CSF) to plasma concentration ratio of atogepant. Healthy adult participants (N = 28) were enrolled into this phase 1, open‐label study and received atogepant 60 mg once daily for 8 days. Steady‐state atogepant concentrations were determined from CSF samples collected on Day 6 at 2
