Introduction of unnatural amino acids into chalcone isomerase
Bioconjugate Chemistry, 1991The active site cysteine residue of chalcone isomerase was rapidly and selectively modified under denaturing conditions with a variety of electrophilic reagents. These denatured and modified enzyme were renatured to produce enzyme derivatives containing a series of unnatural amino acids in the active site.

Site-specific mutagenesis with unnatural amino acids
Trends in Biochemical Sciences, 1989The incorporation of unnatural amino acids into proteins by site-specific mutagenesis provides a valuable new methodology for the generation of novel proteins that possess unique structural and functional features.

Microwave-assisted synthesis of unnatural amino acids
Bioorganic & Medicinal Chemistry Letters, 2008Microwave irradiation has been proven to be a useful tool in the rapid assembly of racemic unnatural amino acids in only two steps. Additional benefits of this methodology are the commercial availability of the inexpensive starting materials and the high yields and high purities of the final amino acid products.

Unnatural Amino Acids in Novel Antibody Conjugates
Future Medicinal Chemistry, 2014Antibody-drug conjugates are an important and emerging drug class for the treatment of cancer. Recent evidence strongly suggests that site-specific drug conjugation results in a homogenous population of molecules with more favorable activity and pharmacokinetic properties than randomly conjugated antibodies.

A Phage Display System with Unnatural Amino Acids
Journal of the American Chemical Society, 2004We report a general method to display peptide-containing unnatural amino acids on filamentous M13 phage. Five distinct unnatural amino acids were site-specifically incorporated at the N-terminal of the M13 phage minor coat protein pIII. Phages that contain p-azidophenylalanine can undergo a highly specific azide-alkyne [3 + 2] cycloaddition reaction ...

Probing Protein Stability with Unnatural Amino Acids
Science, 1992Unnatural amino acid mutagenesis, in combination with molecular modeling and simulation techniques, was used to probe the effect of side chain structure on protein stability. Specific replacements at position 133 in T4 lysozyme included (i) leucine (wt), norvaline, ethylglycine, and alanine to measure the cost of stepwise removal of methyl

Incorporation of unnatural amino acids for synthetic biology
Molecular BioSystems, 2009Abstract The challenge of synthetic biology lies in the construction of artificial cellular systems. This requires the development of modular “parts” that can be integrated into living systems to elicit an artificial, yet programmed, response or function.

IMPAIRMENT OF TUMOR GROWTH BY UNNATURAL AMINO ACIDS
Canadian Journal of Biochemistry and Physiology, 1959Fourteen unnatural amino acids were assayed for their action on the development of a transplantable Novikoff hepatoma tumor, and 10 of them were found to exert some inhibitory effects.

New Developments in the Synthesis of Natural and Unnatural Amino Acids
Annals of the New York Academy of Sciences, 1992SUMMARYAmino acids play an important role in biochemistry and chemistry. They are the building blocks of proteins and play an essential role in the regulation of the metabolism of living organisms. In general, it can be stated that microbial processes (fermentation) are the industrial production methods of choice for large‐scale production of naturally ...

Unnatural Amino Acids for Biological Spectroscopy and Microscopy
Chemical ReviewsDue to advances in methods for site-specific incorporation of unnatural amino acids (UAAs) into proteins, a large number of UAAs with tailored chemical and/or physical properties have been developed and used in a wide array of biological applications. In particular, UAAs with specific spectroscopic characteristics can be used as external reporters to ...
