Structural instability impairs function of the UDP‐xylose synthase 1 Ile181Asn variant associated with short‐stature genetic syndrome in humans

open access: yesFEBS Letters, EarlyView.
The Ile181Asn variant of human UDP‐xylose synthase (hUXS1), associated with a short‐stature genetic syndrome, has previously been reported as inactive. Our findings demonstrate that Ile181Asn‐hUXS1 retains catalytic activity similar to the wild‐type but exhibits reduced stability, a looser oligomeric state, and an increased tendency to precipitate ...

Cell wall target fragment discovery using a low‐cost, minimal fragment library

open access: yesFEBS Letters, EarlyView.
LoCoFrag100 is a fragment library made up of 100 different compounds. Similarity between the fragments is minimized and 10 different fragments are mixed into a single cocktail, which is soaked to protein crystals. These crystals are analysed by X‐ray crystallography, revealing the binding modes of the bound fragment ligands.

Some cone separation results and applications

open access: yesJournal of Numerical Analysis and Approximation Theory, 2008
In this note we present some cone separation results in infinite dimensional spaces. Our approach is mainly based on two different types of cone outer approximation. Then we consider an application to vector optimization.

Calpain small subunit homodimerization is robust and calcium‐independent

open access: yesFEBS Letters, EarlyView.
Calpains dimerize via penta‐EF‐hand (PEF) domains. Using single‐molecule force spectroscopy, we measured the strength and kinetics of PEF–PEF homodimer binding. The interaction is robust, shows a transient conformational step before dissociation, and remains largely insensitive to Ca2+.

Vector‐valued optimal Lipschitz extensions [PDF]

open access: yesCommunications on Pure and Applied Mathematics, 2011
AbstractConsider a bounded open set $\font\open=msbm10 at 10pt\def\R{\hbox{\open R}}U \subset \R^n$ and a Lipschitz function $\font\open=msbm10 at 10pt\def\R{\hbox{\open R}}g: \partial U \to \R^m$. Does this function always have a canonical optimal Lipschitz extension to all of U? We propose a notion of optimal Lipschitz extension and address existence

A methionine‐lined active site governs carbocation stabilization and product specificity in a bacterial terpene synthase

open access: yesFEBS Letters, EarlyView.
This study reveals a unique active site enriched in methionine residues and demonstrates that these residues play a critical role by stabilizing carbocation intermediates through novel sulfur–cation interactions. Structure‐guided mutagenesis further revealed variants with significantly altered product profiles, enhancing pseudopterosin formation. These
