Establishment of a humanized patient‐derived xenograft mouse model of high‐grade serous ovarian cancer for preclinical evaluation of combination immunotherapy

open access: yesMolecular Oncology, EarlyView.
We have established a humanized orthotopic patient‐derived xenograft (Hu‐oPDX) mouse model of high‐grade serous ovarian cancer (HGSOC) that recapitulates human tumor–immune interactions. Using combined anti‐PD‐L1/anti‐CD73 immunotherapy, we demonstrate the model's improved biological relevance and enhanced translational value for preclinical ...

Hippo pathway at the crossroads of stemness and therapeutic resistance in breast cancer

open access: yesMolecular Oncology, EarlyView.
Dysregulation of the Hippo pathway drives nuclear accumulation of YAP/TAZ, activating stemness‐related transcriptional programs that sustain breast cancer stemness and fuel therapeutic resistance across subtypes, underscoring Hippo signaling as a targetable vulnerability. Figure created and edited with BioRender.com.

Deciphering transcriptional plasticity in pancreatic ductal adenocarcinoma reveals alterations in sensory neuron innervation

open access: yesMolecular Oncology, EarlyView.
Pancreatic sensory neurons innervating healthy and PDAC tissue were retrogradely labeled and profiled by single‐cell RNA sequencing. Tumor‐associated innervation showed a dominant neurofilament‐positive subtype, altered mitochondrial gene signatures, and reduced non‐peptidergic neurons.

Longitudinal circulating tumor DNA profiling in patients with advanced endometrial cancer using an off‐the‐shelf targeted NGS panel

open access: yesMolecular Oncology, EarlyView.
Intratumour heterogeneity complicates precision management of advanced endometrial cancer. Circulating tumor DNA (ctDNA) offers a minimally invasive strategy to capture tumor evolution and therapeutic resistance. Here, we compare tumor‐agnostic NGS with tumor‐informed ddPCR, outlining their relative sensitivity, concordance, and clinical implications ...

Visual detection of Coxsackievirus A6 using a reverse transcription polymerase spiral reaction method

open access: yesFrontiers in Cellular and Infection Microbiology
Coxsackievirus A6 (CVA6) ranks as a primary enterovirus associated with hand-foot-mouth disease (HFMD) and herpangina (HA). Given its significant role in these diseases, there is an urgent need for an efficient identification method.

Aging and visual pattern detection

open access: yesJournal of Vision
A single psychophysical experiment evaluated observers' ability to detect visual patterns embedded in noise; effects of stimulus complexity and observer age were also evaluated. Eighteen younger and older observers participated in the experiment (mean ages were 20.3 and 72.6 years, respectively).

Interpreting the effects of DNA polymerase variants at the structural level

open access: yesMolecular Oncology, EarlyView.
Using MAVISp and molecular dynamics simulations, we analyzed over 60 000 missense variants in POLE and POLD1 from ClinVar, COSMIC, cBioPortal, and saturation mutagenesis. Identified mechanistic indicators, including stability, binding, and long‐range, enable structural interpretation, providing ACMG‐like evidence for possible reclassification of VUS ...

Design of an intermittent dust removal system for visual detection of longitudinal tearing of coal mine conveyor belts

open access: yesGong-kuang zidonghua
In the application of visual detection of longitudinal tearing of coal mine conveyor belts, the visual system is prone to problems such as dust blockage and contamination, which affect the clarity and accuracy of image acquisition.

Finding novel vulnerabilities of hypomorphic BRCA1 alleles

open access: yesMolecular Oncology, EarlyView.
Synthetic lethality screens performed to identify novel vulnerabilities often model complete gene loss, thereby overlooking patient‐derived hypomorphic mutations. In this study, we have performed genome‐wide CRISPR screens on BRCA1 hypomorphic mutations, showing BRCA1I26A behaves like wild‐type, while BRCA1R1699Q mimics deficiency. Furthermore, we have

Design and experiment of miss-seeding detection and preparatory seed scraper-belt compensation mechanism based on improved YOLOv5s for potato seed-metering devices

open access: yesFrontiers in Plant Science
This study addresses the issue of miss seeding in spoon-chain potato seed-metering devices, which impacts planting efficiency and quality, by proposing a miss-seeding detection and compensation system based on an improved YOLOv5s model integrated with a ...
