Finding novel vulnerabilities of hypomorphic BRCA1 alleles

open access: yesMolecular Oncology, EarlyView.
Synthetic lethality screens performed to identify novel vulnerabilities often model complete gene loss, thereby overlooking patient‐derived hypomorphic mutations. In this study, we have performed genome‐wide CRISPR screens on BRCA1 hypomorphic mutations, showing BRCA1I26A behaves like wild‐type, while BRCA1R1699Q mimics deficiency. Furthermore, we have

First x-ray nanoimaging experiments at NanoMAX

open access: yes, 2017
NanoMAX is a hard x-ray nanoimaging beamline at the new Swedish synchrotron radiation source MAX IV that became operational in 2016. Being a beamline dedicated to x-ray nanoimaging in both 2D and 3D, NanoMAX is the first to take full advantage of MAX IVs

A light‐triggered Time‐Resolved X‐ray Solution Scattering (TR‐XSS) workflow with application to protein conformational dynamics

open access: yesFEBS Open Bio, EarlyView.
Time‐resolved X‐ray solution scattering captures how proteins change shape in real time under near‐native conditions. This article presents a practical workflow for light‐triggered TR‐XSS experiments, from data collection to structural refinement. Using a calcium‐transporting membrane protein as an example, the approach can be broadly applied to study ...

Analysing the significance of small conformational changes and low occupancy states in serial crystallographic data

open access: yesFEBS Open Bio, EarlyView.
This protocol paper outlines methods to establish the success of a time‐resolved serial crystallographic experiment, by means of statistical analysis of timepoint data in reciprocal space and models in real space. We show how to amplify the signal from excited states to visualise structural changes in successful experiments.

Enhanced colour encoding of materials discrimination information for multiple view dual-energy x-ray imaging

open access: yes, 2009
This thesis reports an investigation into dual-energy X-ray discrimination techniques. These techniques are designed to provide colour-coded materials discrimination information in a sequence of perspective images exhibiting sequential parallax.

Proteasomal degradation of intracellularly expressed Amblyomin‐X limits suicide gene therapy potential in melanoma cells

open access: yesFEBS Open Bio, EarlyView.
This study explores the feasibility of expressing the antitumoral protein Amblyomin‐X through a suicide gene therapy approach and investigates its intracellular fate after gene delivery. Although the gene is efficiently expressed, melanoma cells rapidly degrade the Amblyomin‐X protein via proteasome activity.

Novel spectral-timing methods for X-ray binary variability studies

open access: yes, 2012
This work explores the connection between the spectral properties of the X-ray emission produced by black hole X-ray binaries (BHXRBs) and the variability properties of these sources that are embedded in light curves.

Evolutionarily divergent DUF4465 domains have a common vitamin B12‐binding function

open access: yesFEBS Open Bio, EarlyView.
We show that DUF4465 family proteins, widespread across bacteria from gut microbiomes, hydrothermal vents, and soil, share a common vitamin B12‐binding function. These augmented β‐jellyroll proteins bind vitamin B12 via extended loops. Our findings establish sequence‐diverse DUF4465 proteins as a widespread class of B12‐binding proteins, highlighting ...

X-ray scatter correcting methods for digital radiographic imaging

open access: yesJournal of Associated Medical Sciences, 2018
X-ray scattering correction method has been the primary means of enhancing radiographic images for quite some time. X-ray scattering is major deterioration factor that decreases image contrast and increases the image granularity in a radiographic image ...

Two methods for separating the magnetospheric solar wind charge exchange soft X-ray emission from the diffuse X-ray background

open access: yesEarth and Planetary Physics
Solar wind charge exchange (SWCX) is the process of solar wind high-valence ions exchanging charges with neutral components and generating soft X-rays. Recently, detecting the SWCX emission from the magnetosphere is proposed as a new technique to study ...
