Detection of homologous recombination between yeast artificial chromosomes with overlapping inserts

open access: yesNucleic Acids Research, 1991
We have developed a system which facilitates the detection of recombination between Yeast Artificial Chromosomes (YAC's) carrying homologous inserts. The system consists of a classical YAC vector, a new YAC vector and two appropriately labelled yeast strains of opposite mating type.

Transposons as Tools for Future Genome Engineering in Yeasts and Filamentous Fungi

open access: yesYeast, EarlyView.
Applications of transposon‐based tools for genome engineering in fungi include transposon‐mediated mutagenesis and targeted genomic integration. ABSTRACT Transposons are fundamental genetic elements that have profoundly shaped the architecture of eukaryotic genomes.

Quantifying and resolving multiple vector transformants in S. cerevisiae plasmid libraries

open access: yesBMC Biotechnology, 2009
Background In addition to providing the molecular machinery for transcription and translation, recombinant microbial expression hosts maintain the critical genotype-phenotype link that is essential for high throughput screening and recovery of proteins ...

CRISPR-PCDup: a novel approach for simultaneous segmental chromosomal duplication in Saccharomyces cerevisiae

open access: yesAMB Express, 2020
In our previous study, a novel genome engineering technology, PCR-mediated chromosome duplication (PCDup), was developed in Saccharomyces cerevisiae that enabled the duplication of any desired chromosomal region, resulting in a segmental aneuploid.

Identification of Yeast Protein Sequences With Similarity to the ESCRT‐III Protein Snf7

open access: yesYeast, EarlyView.
ABSTRACT Endosomal sorting complex required for transport (ESCRT‐III) is a membrane remodeling complex involved in a large number of cellular processes. It appears to perform an essential function in eukaryotes, since to date no eukaryotic organism completely devoid of ESCRT‐III has been found.

Cell cycle-dependent modification of Pot1 and its effects on telomere function [PDF]

open access: yes, 2009
Telomere functions are tightly controlled throughout the cell cycle to allow telomerase access while suppressing a bona fide DNA damage response (DDR) at linear chromosome ends.

Tomato protoplast DNA transformation: physical linkage and recombination of exogenous DNA sequences [PDF]

open access: yes, 1987
Tomato protoplasts have been transformed with plasmid DNA's, containing a chimeric kanamycin resistance gene and putative tomato origins of replication.

CryoBN‐PAGE: A Rapid and Cost‐Effective Method for the Analysis of Yeast Respiratory Complexes and Their Dynamic Organization

open access: yesYeast, EarlyView.
We present cryoBN‐PAGE, a method that enables blue native polyacrylamide gel electrophoresis analysis of mitochondrial respiratory complexes and supercomplexes directly from whole yeast cells using cryogenic milling. This approach eliminates the need for mitochondrial isolation, reducing time and cost while preserving native protein complexes.

DNA polymerase α (swi7) and the flap endonuclease fen1 (rad2) act together in the s-phase alkylation damage response in S. pombe [PDF]

open access: yes, 2012
Polymerase α is an essential enzyme mainly mediating Okazaki fragment synthesis during lagging strand replication. A specific point mutation in Schizosaccharomyces pombe polymerase α named swi7-1, abolishes imprinting required for mating-type switching ...

Yeast Knockout Strain Collection: Driving Functional Genomics, Biotechnological Applications and Human Disease Research

open access: yesYeast, EarlyView.
ABSTRACT The knockout strain collection of Saccharomyces cerevisiae has served as a valuable resource for functional genomics and yeast‐based biotechnology studies. A comprehensive single‐gene knockout strain collection, covering nearly all non‐essential genes, together with complementary mutant collections for essential genes, including temperature ...
