A robust gene-stacking method utilizing yeast assembly for plant synthetic biology
Plant synthetic biology offers the potential to re-engineer crops, but requires efficient methods to prepare constructs for transformation. Here Shih et al. develop jStack, a method that utilizes yeast homologous recombination and a library of DNA parts,

ABSTRACT Allergic diseases arise from aberrant immune recognition of otherwise harmless environmental proteins and are driven by epitope‐specific interactions between allergens and the adaptive immune system. Although component‐resolved diagnostics have improved molecular characterisation of sensitization, they remain limited to whole‐allergen ...

Impact of DNA ligase IV on nonhomologous end joining pathways during class switch recombination in human cells [PDF]
Class switch recombination (CSR) is a region-specific, transcriptionally regulated, nonhomologous recombinational process that is initiated by activation-induced cytidine deaminase (AID).

Mice expressing only mutant PTEN Y138L, a protein which shows normal suppression of cellular AKT yet lacks protein phosphatase activity, die in utero, and heterozygous mice display a range of tumors. This indicates both the lipid and protein phosphatase activities of PTEN work together for normal tumor suppression and embryonic development.

Lego DNA assembling, a simple in vitro method for constructing DNA molecule [PDF]
Digestion-ligation based and recombination based methods for constructing recombinant DNA are the basic techniques in molecular biology and thus built one of the foundations for the modern life sciences. Here we describe a new strategy that can radically

Molecular mechanisms underlying the control of antigenic variation in African trypanosomes [PDF]
African trypanosomes escape the host adaptive immune response by switching their dense protective coat of Variant Surface Glycoprotein (VSG). Each cell expresses only one VSG gene at a time from a telomeric expression site (ES).

The eukaryote‐specific N‐terminal domain (NTD) of eS31 uses two distinct strategies to maintain translation fidelity. During elongation, a positively charged “hotspot” fine‐tunes the selection of incoming aa‐tRNA. During termination, the entire NTD acts as a structural scaffold to ensure the correct positioning of the release factor eRF1.
