HBsAg 重组腺病毒载体的构建、鉴定

【目的】为探索腺病毒载体在基因治疗中的应用及其表达目的抗原的有效性。【方法】用 BamHⅠ 、EcoRⅠ双切 质粒 pBluscript KS+ -HBs, 回收 S 基因片段, 定向克隆插入质粒 pACCMVpL 中相应的酶切位点, 通过快速抽提酶切鉴定筛选 重组质粒。【结果】挑取的 8 个克隆经 BamHⅠ 、EcoRⅠ酶切鉴定, 有 950 bp 的目的基因片段, 证明为重组体。【结论】成功 构建表达 HBsAg 基因的重组腺病毒载体。
