A sensitive and simplified radioimmunoassay for 1,25-dihydroxyvitamin D3

Clinica Chimica Acta, 1984
A sensitive radioimmunoassay system for 1 alpha,25-dihydroxyvitamin D3 [1,25(OH)2D3] with an improved extraction procedure has been developed. Following one-step extraction and prepurification of 1,25(OH)2D3 by 'Extrelut-1' minicolumns final purification was achieved by high-performance liquid chromatography (HPLC) using a radial compression separation

Localized scleroderma-response to 1,25-dihydroxyvitamin D3

Clinical and Experimental Dermatology, 1990
1,25-Dihydroxyvitamin D3 [1,25(OH)2 D3] may be an immunomodulatory drug which could have a role in controlling collagen deposition, and inducing reversal of fibrosis in some tissues. These observations prompted a study of the possible use of this hormone for the treatment of scleroderma.

Immunomodulatory actions of 1,25-Dihydroxyvitamin D3

The Journal of Steroid Biochemistry and Molecular Biology, 1995
The sterol, 1,25-dihydroxyvitamin D3 (1,25(OH)2D3), has immunosuppressive activity. The hormone inhibits the production of lymphokines (IL-2, IFN-gamma) and monocyte-derived cytokine (IL-12) leading to inhibition of helper T cell subset type 1 (Th1).

1,25-Dihydroxyvitamin D3 receptors in mouse colon

Journal of Steroid Biochemistry, 1981
Mouse colon possesses a cytoplasmic receptor-like binder for 1,25-dihydroxy vitamin D3 which appears similar to the receptors previously described in other more classical target organs. The properties of this receptor include a sedimentation coefficient of 3.2S in sucrose gradients prepared in 0.3 M KCl, an equilibrium dissociation constant of 0.2 nM ...

Intestinal absorption of 1,25-dihydroxyvitamin D3 in the rat

American Journal of Physiology-Gastrointestinal and Liver Physiology, 1985
Intestinal absorption of 1,25-dihydroxyvitamin D3 [1,25(OH)2D3] from in vivo jejunal sacs was studied in rats with thoracic duct and bile duct cannulas. In 6 h 86.5% of the administered 1,25(OH)2D3 was absorbed, but only 7.3% was recovered in thoracic duct lymph.

1,25-Dihydroxyvitamin D3 and the regulation of macrophage function

Calcified Tissue International, 1981
Vitamin D3 deficient (D-) mice show a depressed inflammatory response and both inflammatory peritoneal macrophages and bone marrow polymorphonuclear leukocytes of D- mice exhibit a decreased spontaneous migration under agarose. The impaired phagocytic response of peritoneal macrophages from D- mice can be corrected by incubation with 1,25 ...

Immunoregulation by 1,25-dihydroxyvitamin D3: Basic concepts

The Journal of Steroid Biochemistry and Molecular Biology, 2005
1,25-Dihydroxyvitamin D(3) (1,25(OH)(2)D(3)), the biologically active metabolite of Vitamin D(3), not only regulates bone and calcium metabolism but also exerts other biological activities, including immunomodulation via the nuclear Vitamin D receptor expressed in antigen-presenting cells and activated T cells.

Specific binding of 1,25 dihydroxyvitamin D3 in lymphocytes

Metabolism, 1984
We examined ten cellular or tissue sources of lymphocytes for specific binding of 1,25(OH)2D3, the hormonally active form of vitamin D3. A specific-binding protein was found in three of these sources. Scatchard analysis of cytosol from a follicular lymphoma cell line revealed binding sites with a Kd of 7.0 X 10(-11) and a receptor concentration of 6.6 ...

Interactions of 1,25-dihydroxyvitamin D3 and the immune system

Molecular and Cellular Endocrinology, 1985
A series of recent discoveries indicate that the hormonal form of vitamin D3, namely, 1,25(OH)2D3 plays a role in the regulation of the immune system. Cells of the monocyte/macrophage lineage possess receptors for 1,25(OH)2D3 regardless of their activation stage; cells of the lymphoid lineage also express these receptors but only at certain stages of ...

An immunoradiometric assay for 1,25-dihydroxyvitamin D3 receptor

Analytical Biochemistry, 1989
A ligand-independent, quantitative assay has been developed for the measurement of 1,25-dihydroxyvitamin D receptor utilizing purified receptor from pig intestine as a standard and two high affinity monoclonal antibodies directed to two different epitopes on the receptor.
