Synthesis of Multifunctionalized 2,3-Dihydro-4-pyridones and 4-Pyridones via the Reaction of Carbamoylated Enaminones with Aldehydes

The novel and effective diastereoselective synthesis of multifunctionalized dihydropyridones, including CF3-substituted derivatives, has been developed on the basis of the piperidine-promoted domino reaction of carbamoylated enaminones with aldehydes.

Two polymorphs of anhydrous 4-pyridone at 100 K

Two polymorphs of the title compound, C(5)H(5)NO, (I), have been obtained from ethanol. One polymorph crystallizes in the monoclinic space group C2/c [henceforth (I)-M], while the other crystallizes in the orthorhombic space group Pbca [henceforth (I)-O]. In the two forms, the lattice parameters, cell volume and packing motifs are very similar.

Potent antimalarial 4-pyridones with improved physico-chemical properties

Antimalarial 4-pyridones are a novel class of inhibitors of the plasmodial mitochondrial electron transport chain targeting Cytochrome bc1 (complex III). In general, the most potent 4-pyridones are lipophilic molecules with poor solubility in aqueous media and low oral bioavailability in pre-clinical species from the solid dosage form.

Preparation and antiinflammatory activity of 2- and 4-pyridones

Several N-alkyl- and N-arylacetoacetamides have been self-condensed to form pyridones. N-Alkylacetoacetamides give 2-pyridones, while N-arylacetoacetamides give 4-pyridones. In an attempt to develop nonacidic, nonsteroidal antiinflammatory agents, the pyridones were tested in a carrageenan-induced pedal edema assay in rats.

Synthesis of 3-acetyl-2-amino-4-pyridone

A method hxsynthesizing 3-acetyl-2-amino-4-pyyridone has been suggested. The synthesis involves the condensation of dimethylformamide dimethylacetal at the NH2 group of diphenylboron chelate diacetylketene aminal followed by its intramolecular cyclization under the action of NaOMe in MeOH.

Spectrofluorimetric determination of iridium(IV) traces using 4-pyridone derivatives

A new spectrofluorimetric determination of iridium(IV) with 3-hydroxy-2-methyl-1-phenyl-4-pyridone (HX) or 3-hydroxy-2-methyl-1-(4-tolyl)-4-pyridone (HY) is reported. Iridium(IV) react with HX or HY and chelates were extracted into chloroform or dichloromethane. The organic phase showed fluorescence.

A facile synthesis of novel tricyclic 4-pyridones
