A single dose of 0.5 mg pizotifen or a placebo was administered to 10 healthy male volunteers in a double blind cross-over trial. 5-hydroxyindoleacetic acid (5-HIAA) and homovanillic acid (HVA) in hourly urine samples were determined by liquid chromatography with amperometric detection.

The present series of studies of CSF monoamine metabolites support the hypothesis of a role for serotonin in depressive illness, insofar that depressed patients have slightly lower CSF 5-HIAA concentrations than normal controls. Apparently a normal association between CSF 5-HIAA and platelet MAO activity is disrupted in depression.

5-HT and 5-HIAA in cerebrospinal fluid in depression

CSF 5-HT and 5-HIAA were measured in endogenously depressed patients (ICD-9) (n = 23) and controls (n = 11). Distribution of sex, age and body height was similar in the two groups. Non-parametric statistics were used. In depressed patients CSF 5-HT concentrations were found to be higher (P less than or equal to 0.01) than in controls.

Intranasal vasotocin decreases cerebrospinal fluid 5-HIAA levels in man

A single dose (10 ng/kg) of the nonapeptide arginine vasotocin (AVT) administered intranasally to healthy young men, significantly decreased 5-HIAA levels in the lumbar cerebrospinal fluid (CSF) 8 hr after its administration. So far, this represents the smallest amount of an active substance able to alter CSF 5-HIAA levels in man.
