ATF4‐mediated stress response as a therapeutic vulnerability in chordoma

open access: yesMolecular Oncology, EarlyView.
We screened 5 chordoma cell lines against 100+ inhibitors of epigenetic and metabolic pathways and kinases and identified halofuginone, a tRNA synthetase inhibitor. Mechanistically halofuginone induces an integrated stress response, with eIF2alpha phosphorylation, activation of ATF4 and its target genes CHOP, ASNS, INHBE leading to cell death ...

On shortening, lengthening, and accent shifts in Slavic

open access: yesRasprave Instituta za Hrvatski Jezik i Jezikoslovlje, 2017
The paper deals with several problems of Slavic historical accentology – pretonic length in the accentual paradigm c (and b) in South and West Slavic, the neo-circumflex phenomenon (including the accent in the genitive plural), the kȍkōt ‘rooster’ type ...

Indo-Uralic consonant gradation [PDF]

open access: yes, 2010
Koivulehto and Vennemann have recently (1996) revived Posti’s theory (1953) which attributed Finnic consonant gradation to Germanic influence, in particular to the influence of Verner’s law. This theory disregards the major differences between Finnic and

PARP inhibition and pharmacological ascorbate demonstrate synergy in castration‐resistant prostate cancer

open access: yesMolecular Oncology, EarlyView.
Pharmacologic ascorbate (vitamin C) increases ROS, disrupts cellular metabolism, and induces DNA damage in CRPC cells. These effects sensitize tumors to PARP inhibition, producing synergistic growth suppression with olaparib in vitro and significantly delayed tumor progression in vivo. Pyruvate rescue confirms ROS‐dependent activity.

Miscellaneous remarks on Balto-Slavic accentuation [PDF]

open access: yes, 2010
The highly successful conference on Balto-Slavic accentology organized by Mate Kapovic and Ranko Matasovic has given much food for thought. It has clarified the extent of fundamental disagreements as well as established areas of common interest where the

Crucial parameters for precise copy number variation detection in formalin‐fixed paraffin‐embedded solid cancer samples

open access: yesMolecular Oncology, EarlyView.
This study shows that copy number variations (CNVs) can be reliably detected in formalin‐fixed paraffin‐embedded (FFPE) solid cancer samples using ultra‐low‐pass whole‐genome sequencing, provided that key (pre)‐analytical parameters are optimized.

Phenotypic and genotypic characterization of single circulating tumor cells in the follow‐up of high‐grade serous ovarian cancer

open access: yesMolecular Oncology, EarlyView.
Single circulating tumor cells (sCTCs) from high‐grade serous ovarian cancer patients were enriched, imaged, and genomically profiled using WGA and NGS at different time points during treatment. sCTCs revealed enrichment of alterations in Chromosomes 2, 7, and 12 as well as persistent or emerging oncogenic CNAs, supporting sCTC identity.
