Complete nucleotide sequence of the genome of a severe cherry isolate of apple chlorotic leaf spot trichovirus (ACLSV)

The genome of the Balaton 1 severe cherry isolate of apple chlorotic leaf spot trichovirus (ACLSV-Bal1) has been cloned and sequenced. The genomic RNA is 7549 nucleotide long, excluding the poly A tail. The genomic organization, with three overlapping open reading frames (ORF), is similar to that of the other sequenced ACLSV isolates.

Apple chlorotic leaf spot virus (ACLSV) seems to be the causal agent of apricot "viruela" disease. This disease has become an important problem for apricot production in Spain and, for this reason, in the CEBAS-CSIC of Murcia we are setting up a method to evaluate ACLSV resistance.

This study presents the incidence of viruses in cultivated almond in Greece and Cyprus. For this reason a pair of primers was designed to be used for the detection of PNRSV, PDV and ApMV. The aim of this work, was also to study the variablity of the polymerase of PDV and of the capsid protein of ACLSV isolates in almond.

Ιn this study the molecular variability of Apple chlorotic leaf spot virus (ACLSV) in cultivated, ornamental and wild species of the Rosaceae family, was investigated. The serological method ELISA was unreliable and two RT-PCR methods were combined in order to increase its sensitivity.
