BTK inhibitors suppress malignant B‐cell signaling while simultaneously reshaping immune regulation through effects on T cells and the tumor microenvironment. These mechanisms support combination immunotherapy strategies and also underlie the expanding application of BTK inhibitors in solid tumors, autoimmune and inflammatory diseases.

Advancing In Vivo Chimeric Antigen Receptor T‐Cell Engineering to Accelerate Clinical Translation

In vivo CAR‐T cell generation directly engineers endogenous T cells using pre‐manufactured gene delivery vectors that target T cells and deliver CAR‐encoding genetic material. The resulting CAR‐T cells expand in vivo and mediate antigen‐specific cytotoxicity.

Immune‐cell dynamics in the tumor microenvironment converge on four major barriers to effective therapy: failed immune access, defective antigen visibility and priming, loss of effector maintenance, and suppressive tissue remodeling. These barriers emerge from local hypoxic, cytokine, metabolic, and stromal signals.

Overview of the antileukemic effects of Piper nigrum extract in AML. P. nigrum reduced leukemic cell proliferation and glycolytic metabolism through modulation of the PI3K/AKT/mTOR pathway. In vivo, treatment reduced leukemic burden, improved hematological parameters, prolonged survival, and modulated antitumor immune responses.
