A Photoswitchable HaloTag for Spatiotemporal Control of Fluorescence in Living Cells

open access: yesAngewandte Chemie, EarlyView.
Photoswitchable fluorophores are critical for advanced bioimaging. Here, we develop a photoswitchable self‐labeling HaloTag that can reversibly modulate the emission of a bound fluorogenic dye via a light‐induced conformational change. In living cells, this tunable chemigenetic system enables precise spatiotemporal activation and control of emitter ...

Cupin‐Type Dimethylsulfoniopropionate Lyase from Pelagibacter ubique (DddKPu) Catalyzes Aza‐Michael Addition of Primary and Secondary Amines to Acrylic Acid

open access: yesAngewandte Chemie, EarlyView.
A dimethylsulfoniopropionate lyase (DddK) was identified as an efficient aza‐Michaelase for the addition of amines to acrylate. Screening 74 amines with wild‐type DddK and its W26G variant yielded N‐substituted and N,N‐disubstituted β‐amino acids in 30%–93% isolated yield.

On‐Target Photoassembly of Pyronin Dyes for Super‐Resolution Microscopy

open access: yesAngewandte Chemie, EarlyView.
We report a new class of diaryl ether and diaryl silane compounds bearing hydrazone substituents that, upon irradiation, cyclize to a fluorescent (silicon−)pyronin dye. After optimizing their photophysical properties, we introduced reactive tags into their structure for biological labeling, and demonstrated their utility in STED, PALM, and MINFLUX ...

Reduced Pressure Ionization Enhances Native Mass Spectrometry of Proteins and Protein Complexes

open access: yesAngewandte Chemie, EarlyView.
Reduced pressure for electrospray ionization of native proteins narrows peak widths and increases signal. This straightforward add‐on to existing native MS platforms improves performance in challenging buffers and enables detection of protein interactions at concentrations as low as 50 nM, without significantly disrupting structure as confirmed by ...

Self‐Assembly, Rearrangement, and Disassembly of {Cr6} Horseshoe Oligomers

open access: yesAngewandte Chemie, EarlyView.
Using a combination of ion mobility mass spectrometry (IM‐MS), tandem mass spectrometry (MS2), and low‐temperature scanning tunneling microscopy (STM), we elucidate the self‐assembly, rearrangement, and disassembly processes of polymetallic {Cr6}x (x = 1 – 5) horseshoe oligomers.
