Immunological Outcomes of Allergen-Specific Immunotherapy in Food Allergy

open access: yesFrontiers in Immunology, 2020
IgE-mediated food allergies are caused by adverse immunologic responses to food proteins. Allergic reactions may present locally in different tissues such as skin, gastrointestinal and respiratory tract and may result is systemic life-threatening ...

Major Cat Allergen Fel d 4: Structure and Identification of a Cross‐Reactive IgE‐Epitope‐Containing Area

open access: yesAllergy, EarlyView.
This study reports the three‐dimensional structure of an important cat allergen, Fel d 4, that shares high homology with Equ c 1 (i.e., major horse allergen) and Can f 6 (i.e., dog allergen). The C‐terminal region of the Fel d 4 contains a major IgE‐reactive epitope.

Long‐Term Effect of Allergen Immunotherapy in Responder Local Allergic Rhinitis Patients: Symptom Control, and Prevention of Asthma and Allergic Sensitizations

open access: yesAllergy, EarlyView.
LAR patients who respond to AIT during the first year and are treated for two more years continue improving (less symptoms and medication intake) during the following seven years. AIT prevents the onset of asthma and allergic sensitizations in responder LAR individuals during the seven years following discontinuation.

Novel monoclonal antibodies against house dust mite allergen Der p 21 and their application to analyze allergen extracts [PDF]

open access: yesPeerJ
Background Allergen extracts and recombinant allergens are used in allergy diagnostics and immunotherapy. Since allergen extracts from different manufacturers lack proper standardization regarding their composition, monoclonal antibodies (MAbs) against ...

A Straightforward Algorithm for Diagnosing Hymenoptera Venom Allergy and Identifying the Relevant Venom for Immunotherapy

open access: yesAllergy, EarlyView.
Conventional tests, such as specific IgE (sIgE) or intradermal testing (IDT), are usually sufficient. Combining both tests increases sensitivity, while additional tests, like the basophil activation test (BAT), provide no further benefit for the majority of patients.

Two years of treatment with the recombinant grass pollen allergy vaccine BM32 induces a continuously increasing allergen-specific IgG4 response

open access: yesEBioMedicine, 2019
Background: BM32, a grass pollen allergy vaccine containing four recombinant fusion proteins consisting of hepatitis B-derived PreS and hypoallergenic peptides from the major timothy grass pollen allergens adsorbed on aluminium hydroxide has been shown ...

Decreased Severity of Oral Immunotherapy Induced Gastrointestinal Eosinophilic Reactions Following Revisions in Milk‐Treatment Protocol

open access: yesAllergy, EarlyView.
A revised milk OIT protocol with lower daily allergen load and slower dose escalation reduced the severity and delayed onset of OIT‐induced gastrointestinal eosinophilic reactions (OITIGER). Elevated baseline absolute eosinophil count (AEC > 600/μL) was a consistent independent risk factor for OITIGER, while older age was protective.

Practical Recommendations for Mixing Allergy Immunotherapy Extracts

open access: yesAllergy & Rhinology, 2015
Critical aspects of formulating allergy immunotherapy vaccines include the selection, total number, and proportions of each allergen component in therapeutic mixtures.

Oral Immunotherapy Induces Shift in lncRNA Expression Modulating Allergen‐Specific Immune Responses

open access: yesAllergy, EarlyView.
We used PBMC transcriptome from 50 hen's egg‐allergic patients to study the role of lncRNA in OIT. Positive correlation was found between 17 lncRNAs and the IL‐4/IL‐13 pathway in vivo, and the expression of the selected lncRNA aligned with CXCL8 upon PBMC egg‐restimulation.

Basophil Activation Test as Biomarker of Severity and Threshold of Allergic Reactions to Cow's Milk During Oral Food Challenges

open access: yesAllergy, EarlyView.
The threshold of reactivity during fresh milk challenges was 70% lower than that of baked milk challenges, reflecting the higher allergenicity of fresh milk. BAT was the only biomarker statistically significantly different between severity and threshold groups, for baked milk and fresh milk allergies, respectively.
