Amrinone metabolism

Clinical Pharmacology and Therapeutics, 1981
High-performance liquid chromatographic methods for the analysis of amrinone in plasma and for both amrinone and its N-acetyl metabolite in urine were developed and applied to measure specimens obtained from a number of healthy men who had received intravenous or oral amrinone. The intravenous doses ranged from 0.8 to 2.2 mg/kg. Terminal elimination of

Dose proportionality of amrinone

Clinical Pharmacology and Therapeutics, 1983
Amrinone was given to 18 healthy subjects in doses of 75, 150, and 225 mg in a randomized crossover design. Plasma levels were shown to rise in proportion to dose. The mean plasma AUC, extrapolated to infinite time, was determined for each dose level; the values obtained were 4, 8.18, and 12.35 micrograms .

Electrophysiologic effects of amrinone

The American Journal of Cardiology, 1985
Patients with congestive heart failure (CHF) have a high prevalence of complex ventricular arrhythmias. Accordingly, the electrophysiologic effects of new drugs for the treatment of CHF should be studied to determine whether they are safe in this population of patients.

Amrinone: pharmacokinetics and pharmacodynamics

Journal of Cardiothoracic Anesthesia, 1989
Amrinone is a selective inhibitor of phosphodiesterase fraction 3 in both cardiac and smooth muscle. Intravenous administration in humans produces increased contractility and vasodilation of venous capacitance and arterial conductance vessels. The elimination half-life in healthy patients is reported to be 2.6 to 4.1 hours, making it the only long ...

Amrinone‐Mediated Thrombocytopenia

Scandinavian Journal of Haematology, 1983
Amrinone, a new cardioactive drug, was administered for, on average, 27.9 d to 12 patients in severe congestive heart failure. Thrombocytopenia occurred in 4 patients, while a progressive decline in platelet numbers developed in 3 additional patients.
