SAR228810: an antibody for protofibrillar amyloid β peptide designed to reduce the risk of amyloid-related imaging abnormalities (ARIA)

open access: yesAlzheimer’s Research & Therapy, 2018
Background Anti-amyloid β (Aβ) immunotherapy represents a major area of drug development for Alzheimer’s disease (AD). However, Aβ peptide adopts multiple conformations and the pathological forms to be specifically targeted have not been identified.

Targeting Hippocampal PTEN Suppresses Ferroptosis and Rescues Cognitive Decline in Alzheimer's Disease via Dual AKT/GSK3β/Nrf2 and AKT/STAT3 Axes

open access: yesAdvanced Science, EarlyView.
PTEN knockdown activates AKT phosphorylation, promoting Nrf2 nuclear translocation and STAT3 activation, which upregulates GPX4 and antioxidant enzymes HO‐1, SOD1, SOD2, and NQO1. These coordinated changes reduce lipid peroxidation and ROS, inhibit ferroptosis, and ultimately ameliorate cognitive impairment in APP/PS1 transgenic mice, highlighting a ...

Amyloid-Related Imaging Abnormalities in Anti-Amyloid Monoclonal Antibody Therapy for Alzheimer’s Disease: Expert Recommendation for Standard MRI Protocol

open access: yesJournal of the Korean Society of Radiology
The introduction of anti-amyloid therapies for Alzheimer’s disease (AD), such as lecanemab (Lequembi ®), which was recently approved in Korea, necessitates careful monitoring for amyloid-related imaging abnormalities (ARIA) using brain MRI. To optimize

Entorhinal Astrocyte Transplants Restore Spatial Exploration and Alleviate Amyloid‐Beta Pathology in Alzheimer's Mice

open access: yesAdvanced Science, EarlyView.
Impaired MEC astrocytic Ca2+ signaling is associated with fragmented spatial exploration in AD mice. Region‐specific glial progenitor transplantation generates engrafted astrocytes that are accompanied by improved AQP4 polarization, reduced amyloid‐β‐associated pathology, attenuated neuroinflammation, preserved synaptic integrity, and ameliorated ...

Imaging microglial activation and glucose consumption in a mouse model of Alzheimer's disease. [PDF]

open access: yes, 2012
In Alzheimer’s disease (AD), persistent microglial activation as sign of chronic neuroinflammation contributes to disease progression. Our study aimed to in vivo visualize and quantify microglial activation in 13- to 15-month-old AD mice using [11C]-(R ...

Phase Separation, Material State, and Condensate Fate in Mammalian Autophagy

open access: yesAdvanced Science, EarlyView.
ABSTRACT Biomolecular phase separation has emerged as a key organizing principle in macroautophagy (hereafter autophagy). In mammalian cells, phase‐separated condensates not only serve as substrates for selective degradation, but also act as dynamic platforms for cargo recognition, signaling integration, and autophagosome assembly.

Early Spatial and Contextual Coding Deficits in Hippocampal CA1 Precede Performance Decline in an Alzheimer's Disease Model

open access: yesAdvanced Science, EarlyView.
Two‐photon calcium imaging in behaving 5xFAD mice reveals early CA1 coding deficits before behavioral decline. Task‐related place cell activity and splitter cell trajectory coding are impaired, with reduced retrospective dominance and greater prospective weighting.

Tau Aggregate Imaging and Transcriptomics of Alzheimer's Disease Brain at Different Stages of Disease

open access: yesAdvanced Science, EarlyView.
The protein aggregates and gene expression in the middle temporal gyrus (MTG) and somatosensory cortex (SOM) of the postmortem brains of 13 Alzheimer's disease patients were studied in detail, revealing that small hyperphosphorylated tau aggregates increase with Braak stage driven by microglial inflammation.
