Control of mitotic transitions by the anaphase–promoting complex [PDF]

open access: yesPhilosophical Transactions of the Royal Society of London. Series B: Biological Sciences, 1999
Proteolysis controls key transitions at several points in the cell cycle. In mitosis, the activation of a large ubiquitin–protein ligase, the anaphase–promoting complex (APC), is required for anaphase initiation and for exit from mitosis. We show that APC is under complex control by a network of regulatory factors, CDC20, CDH1 and MAD2.

Securin Is Not Required for Chromosomal Stability in Human Cells [PDF]

open access: yes, 2005
Abnormalities of chromosome number are frequently observed in cancers. The mechanisms regulating chromosome segregation in human cells are therefore of great interest.

The Anaphase-promoting Complex Coordinates Initiation of Lens Differentiation [PDF]

open access: yesMolecular Biology of the Cell, 2007
Lens development requires the precise coordination of cell division and differentiation. The mechanisms by which the differentiation program is initiated after cell cycle arrest remains not well understood. Cyclin-dependent kinase inhibitors (CKIs), such as p15 and p21, have been suggested to be critical components that inhibit G1 progression and ...

Regulation of OLC1 protein expression by the anaphase-promoting complex. [PDF]

open access: yesOncol Lett, 2019
Overexpressed in lung cancer 1 (OLC1) is a potential oncogene overexpressed in human lung cancer and in other types of malignant tumor. The elevated expression of OLC1 contributes to tumor genesis and progression. However, the mechanisms regulating the expression of OLC1 remain unclear. In the present study, using lung and esophageal cancer cell lines,

Survivin and Aurora Kinase A control cell fate decisions during mitosis

open access: yesMolecular Oncology, EarlyView.
Aurora A interacts with survivin during mitosis and regulates its centromeric role. Loss of Aurora A activity mislocalises survivin, the CPC and BubR1, leading to disruption of the spindle checkpoint and triggering premature mitotic exit, which we refer to as ‘mitotic slippage’.

Timed degradation of Mcl-1 controls mitotic cell death [PDF]

open access: yes, 2018
Mitotic arrest can result in cell death through the process of apoptosis. We have shown by live-cell imaging that the ubiquitin-proteasome dependent proteolysis of the apoptotic regulator Mcl-1 under the control of the anaphase-promoting complex or ...

BMI‐1 modulation and trafficking during M phase in diffuse intrinsic pontine glioma

open access: yesFEBS Open Bio, EarlyView.
The schematic illustrates BMI‐1 phosphorylation during M phase, which triggers its translocation from the nucleus to the cytoplasm. In cycling cells, BMI‐1 functions within the PRC1 complex to mediate H2A K119 monoubiquitination. Following PTC596‐induced M phase arrest, phosphorylated BMI‐1 dissociates from PRC1 and is exported to the cytoplasm via its

Novel functions for the anaphase-promoting complex in neurobiology [PDF]

open access: yesSeminars in Cell & Developmental Biology, 2011
In recent years, diverse and unexpected neurobiological functions have been uncovered for the major cell cycle-regulated ubiquitin ligase, the anaphase-promoting complex (APC). Functions of the APC in the nervous system range from orchestrating neuronal morphogenesis and synapse development to the regulation of neuronal differentiation, survival, and ...

Phase‐Separation of YAP Mediates AJUBA Super Enhancer Activation to Promote Aberrant Mitosis in Breast Cancer

open access: yesAdvanced Science, EarlyView.
YAP undergoes phase separation to activate the AJUBA super‐enhancer, driving aberrant mitosis through spindle checkpoint dysregulation in breast cancer. This study uncovers a mechanistic link between YAP condensates, super‐enhancer activation, and aneuploidy formation.

Mechanisms of ubiquitin transfer by the anaphase-promoting complex [PDF]

open access: yesJournal of Biology, 2009
The anaphase-promoting complex (APC) is a ubiquitin-protein ligase required for the completion of mitosis in all eukaryotes. Recent mechanistic studies reveal how this remarkable enzyme combines specificity in substrate binding with flexibility in ubiquitin transfer, thereby allowing the modification of multiple lysines on the substrate as well as ...
