Androgen Deprivation Therapy in High-Risk Localized and Locally Advanced Prostate Cancer
The recommended treatment for high-risk localized or locally advanced prostate cancer is radical prostatectomy plus extended pelvic lymph node dissection or radiation therapy plus long-term androgen deprivation therapy. However, some patients are treated

A human 3D bone niche integrating osteoblasts and osteoclasts enables co‐culture with prostate cancer cell lines and patient‐derived organoids. The engineered niche (i) models aspects of reciprocal phenotypic regulation between osteoblasts and cancer cells, (ii) captures osteoclast‐enhanced osteomimicry in tumor cells, and (iii) establishes a ...

This study demonstrates that transcription factor androgen receptor (AR) directly binds the LF promoter, driving lactoferrin overexpression to promote ferritin (FTH1/FTL) upregulation and inhibit p53‐ALOX12‐mediated ferroptosis in prostate cancer. Lactoferrin could be a new potential therapeutic target in prostate cancer.

Androgen Receptor Signaling in the Development of Castration-Resistant Prostate Cancer
Most prostate cancers are androgen-sensitive malignancies whose growths depend on the transcriptional activity of the androgen receptor (AR). In the 1940s, Charles Huggins demonstrated that the surgical removal of testes in men can result in a dramatic ...

Denosumab in men receiving androgen-deprivation therapy for prostate cancer
Androgen-deprivation therapy is well-established for treating prostate cancer but is associated with bone loss and an increased risk of fracture. We investigated the effects of denosumab, a fully human monoclonal antibody against receptor activator of ...

T Cell Exhaustion in Cancer Immunotherapy: Heterogeneity, Mechanisms, and Therapeutic Opportunities
T cell exhaustion limits immunotherapy efficacy. This article delineates its progression from stem‐like to terminally exhausted states, governed by persistent antigen, transcription factors, epigenetics, and metabolism. It maps the exhaustion landscape in the TME and proposes integrated reversal strategies, providing a translational roadmap to overcome
