Coumarin‐Based Aldo‐Keto Reductase Family 1C (AKR1C) 2 and 3 Inhibitors

The AKR1C family of enzymes are overexpressed in numerous cancers where they are involved in drug resistance development. Coumarin amide 3 a possessed IC50 values of 50 nM and 90 nM for AKR1C3 and AKR1C2, respectively, and exhibits ‘drug‐like’ metabolic stability and half‐life in human and mouse liver microsomes and plasma.

Inhibition of the glucocorticoid‐activating enzyme 11β‐hydroxysteroid dehydrogenase type 1 drives concurrent 11‐oxygenated androgen excess

Co‐expression of the glucocorticoid‐activating enzyme HSD11B1 with the androgen‐activating enzyme AKR1C3 in adipose attenuates the biosynthesis of the 11‐oxygenated androgen, 11‐ketotestosterone. HSD11B1 inhibition leads to decreased local glucocorticoid activation but drives concurrent 11‐oxygenated androgen excess.
