2-Pyridyl-thioacetamide (2-PTA) was synthesized at IDR in 1968. This molecule showed strong antiulcer/antisecretory effects in animals and man by inhibiting the proton pump. Based on our work Astra developed omeprazole which is the No. 1 drug in the world from 1996. 2-PTA remained a very popular template for drug design for a quarter of century.

Effect of the enzymatic inhibitor of Kunitz on the gastric lesions from reserpine, from phenylbutazone, from pyloric ligation and by restraint in the rat [PDF]
The protective effects of certain polypeptides on gastric ulcerations caused from reserpine and phenylbutazone in the rate were studied. It was found that the Kunitz enzymatic inhibitor exerts a protective action in regard to gastric lesions.
