Protein Language Model‐Guided Engineering of a 2,3‐Butanediol Dehydrogenase for the Enantioselective Synthesis of Cyclic α‐Hydroxy Ketones

A (2R,3R)‐butanediol dehydrogenase from Bacillus subtilis (BsBDH) is engineered for the enantioselective synthesis of 2‐hydroxycyclohexanone. A PASS computational design strategy is proposed to enhance the thermostability of BsBDH. Moreover, ESM‐1v combined with ISM is utilized for enhancing and inverting its stereoselectivity.

4‐Hydroxybenzyl Alcohol Mitigates Hyperlipidemia‐Associated Depression by Inhibiting Neuroinflammation via the NKIRAS2/NF‐κB Pathway

4‐HBA upregulates NKIRAS2 levels, inhibiting the activation of the NF‐κB pathway and subsequently reducing the levels of neuroinflammatory markers. This modulation helps restore normal mood and behavior in hyperlipidemic conditions, providing a potential therapeutic strategy for managing hyperlipidemia‐associated depression.

H2S‐Releasing Aspirin Nanoparticles Alleviate Endometriosis and Associated Anxiety

Albumin nanoparticles loaded with an H2S‐releasing aspirin derivative (ACS14) hitchhiking neutrophils after intraperitoneal injection and preferentially accumulate in endometriosis lesions in mice. The treatment suppresses lesion growth by regulating PI3K/Akt and reduces inflammation by inhibiting NF‐κB.
