YTHDC1 Orchestrates Glucose and Glutamate Rewiring to Overcome Lethal Metabolic Stress in Triple‐Negative Breast Cancer

open access: yesAdvanced Science, EarlyView.
Lai et al. find that m6A reader YTHDC1 is specifically overexpressed in triple‐negative breast cancer (TNBC) and orchestrates glucose and glutamine metabolism in an m6A‐dependent manner to confer robust adaptability to lethal metabolic stress. Leveraging YTHDC1 knockdown‐induced metabolic vulnerability, an ATF4 mRNA‐targeted nanotherapy is developed to

Slipknotted and unknotted monovalent cation-proton antiporters evolved from a common ancestor.

open access: yesPLoS Computational Biology, 2021
While the slipknot topology in proteins has been known for over a decade, its evolutionary origin is still a mystery. We have identified a previously overlooked slipknot motif in a family of two-domain membrane transporters. Moreover, we found that these

Helicobacter pylori‐Induced Persistent IGF2BP1 Activation Promotes Ferroptosis Resistance in Gastric Tumorigenesis

open access: yesAdvanced Science, EarlyView.
Helicobacter pylori infection induces sustained upregulation of the RNA‐binding protein IGF2BP1 in gastric epithelial cells, establishing a persistent epitranscriptomic program. IGF2BP1 stabilizes SLC7A11 mRNA in an m6A‐dependent manner to suppress ferroptosis and promote epithelial survival under oxidative stress. Pharmacological inhibition of IGF2BP1

Polar or Charged Residues Located in Four Highly Conserved Motifs Play a Vital Role in the Function or pH Response of a UPF0118 Family Na+(Li+)/H+ Antiporter

open access: yesFrontiers in Microbiology, 2020
Functionally uncharacterized UPF0118 family has been re-designated as autoinducer-2 exporter (AI-2E) family since one of its members, Escherichia coli YdgG, was identified to function as an AI-2E.

Design of a Novel Diamine Biosensor to Guide the Engineering of Escherichia coli for Cadaverine Overproduction

open access: yesAdvanced Science, EarlyView.
A high‐throughput screening platform was established by integrating an in vivo cadaverine biosensor with a fluorescence‐activated cell sorting (FACS) system. Biosensor‐driven specific exporter tuning, pathway optimization and genome‐wide target mining were conducted to maximize cadaverine production in Escherichia coli.

Dual Repression by IPA1 Fine‐Tunes OsbZIP79‐Mediated Salt Tolerance in Rice

open access: yesAdvanced Science, EarlyView.
Dual repression by IPA1 fine‐tunes OsbZIP79‐mediated salt tolerance in rice: direct transcriptional inhibition under normal conditions and salt‐induced degradation under stress. This dual mechanism activates OsbZIP79 to regulate Na+/K+ homeostasis and redox balance via downstream genes, enabling optimal salt stress response.

Characterization of a novel two-component Na+(Li+, K+)/H+ antiporter from Halomonas zhaodongensis

open access: yesScientific Reports, 2017
In this study, genomic DNA was screened for novel Na+/H+ antiporter genes from Halomonas zhaodongensis by selection in Escherichia coli KNabc lacking three major Na+/H+ antiporters.

Cation Antiports of Animal Plasma Membranes [PDF]

open access: yesJournal of Experimental Biology, 1994
ABSTRACT Both organic and inorganic solutes can be transported across biological membranes against their electrochemical gradient, by coupling to the downhill movement of another species. In animal cells, the underlying mechanism most often involves symport (cotransport) or antiport (countertransport) with either sodium or hydrogen ions.
